Buffer before Lyophilization

Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.

Function
Major component of the type IV pilus (T4P) that plays a role in surface and host cell adhesion, colonization, biofilm maturation, virulence, and twitching, a form of surface-associated motility facilitated by cycles of extension, adhesion, and retraction of T4P fibers. In addition, plays a critical role in type II secretion of exoenzymes by interacting with specific components such as XcpT or XcpU. Modulates host calcium signaling through interaction with host calcium-modulating cyclophilin ligand.

  1. Glycosylation of type 4 fimbrial protein (PilA) protect Pseudomonas aeruginosa from certain pilus-specific phages. Alterations to PilA sequence can block phage infection, but glycosylation is less likely to create disadvantageous phenotypes. Through construction of chimeric phages, specific phage tail proteins allow infection of P. aeruginosa strains with glycosylated pili. PMID: 29133883
  2. PilS's phosphatase motif was required for the autoregulation of pilA transcription, suggesting that under conditions where PilA is abundant, the PilA-PilS interaction promotes PilR dephosphorylation and thus down-regulation of further pilA transcription. PMID: 27162347
