Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Regulatory subunit of the calcium activated potassium KCNMA1 (maxiK) channel. Modulates the calcium sensitivity and gating kinetics of KCNMA1, thereby contributing to KCNMA1 channel diversity. Increases the apparent Ca(2+)/voltage sensitivity of the KCNMA1 channel. It also modifies KCNMA1 channel kinetics and alters its pharmacological properties. It slows down the activation and the deactivation kinetics of the channel. Acts as a negative regulator of smooth muscle contraction by enhancing the calcium sensitivity to KCNMA1. Its presence is also a requirement for internal binding of the KCNMA1 channel opener dehydrosoyasaponin I (DHS-1) triterpene glycoside and for external binding of the agonist hormone 17-beta-estradiol (E2). Increases the binding activity of charybdotoxin (CTX) toxin to KCNMA1 peptide blocker by increasing the CTX association rate and decreasing the dissociation rate.
Gene References into Functions
  1. Study showed that in human and rat arterial myocytes, membrane depolarization rapidly increased the cell surface abundance of auxiliary BK beta1 subunits but not that of the pore-forming BKalpha channels. Membrane depolarization stimulated voltage-dependent Ca(2+) channels, leading to Ca(2+) influx and the activation of Rho kinase (ROCK) 1 and 2. PMID: 28487419
  2. n-3 PUFAs protect the coronary BK channel function and coronary vasoreactivity in diabetic rats as a result of not only increasing BK-beta1 protein expressions, but also decreasing coronary artery tension and coronary smooth muscle cytosolic Ca2+ concentrations. PMID: 29040972
  3. Data show that leukotriene B4, but not other leukotrienes of the LT4 family, activates recombinant (cbv1-beta1) Lrge-conductance calcium-activated potassium (BK) channels. PMID: 25371198
  4. aging decreases capability of MaxiK channel in regulating vascular tone in the MA, which may be partially mediated by unparallel downregulation of alpha- and beta1-subunit expression PMID: 24051206
  5. The expression of BK(Ca) beta1- but not alpha-subunit was significantly enhanced after exercise training PMID: 22660813
  6. BKbeta1 transcripts and protein are detected in several astrocytic populations and cultured cells, to a lesser degree than BKbeta4 channels. PMID: 21438011
  7. KCNMB1 was the dominant beta subunit in the ductus arteriosus, and the amount of beta1 mRNAs was greatest in the mature DA. PMID: 20936291
  8. Immunocytochemical staining of rat hypothalamic neurons indicates strong BKbeta1 expression in the supraoptic nucleus and the magno- and parvocellular parts of the paraventricular nucleus. PMID: 20443052
  9. ageing was associated with a reduction in BK(Ca) channels, and exercise training (ET) partly reversed this reduction. low-intensity ET may be beneficial in restoring age-related decline in coronary vasodilatory properties mediated by BK(Ca) channels. PMID: 20139169
  10. Downregulation of BKCa channel beta1-subunit in the type-2 diabetic cerebral artery smooth muscle cells contributes to cerebral vascular complication in type-2 diabetes via reducing activity of BKCa channels, increasing vascular tone and blood pressure. PMID: 20334613
  11. Kvbeta 1.1 contributes to the developmental control of the A-typepotassium channel current in hippocampal neurons. PMID: 12890510
  12. lower expression of the beta1 subunit during genetic borderline and severe hypertension reduced BK channel activity by decreasing the sensitivity of these channels to physiological changes in Ca2+ PMID: 14551242
  13. activation of specific PKC isozymes increases BK(Ca)-channel activity in Sprague-Dawley rat PASMC via cyclic GMP-dependent protein kinase, which suggests a unique signaling mechanism for vasodilatation. PMID: 14966080
  14. Increased BK beta1 expression in mesenteric arteries may represent a compensatory reaction to limit the development of hypertension. PMID: 16814121
  15. DiBAC(4)(3) activated whole-cell BKalphabeta1 and BKalphabeta4 currents in the same concentration range but partially blocked BKalphabeta2 currents. Structure activity relationships are discussed. PMID: 17209121
  16. The large conductance (BK) beta 1 channel subunit confers Ca2+-sensitivity to BK channel complexes, and both transcript and protein levels for BK beta 1 were appreciably lower in diabetic retinal arterioles. PMID: 17293477
  17. There is a selective distribution of auxiliary beta1 subunits to the somatic and dendritic compartments and beta4 subunits to the terminal compartment of neurons in the hypothalamic-neurohypophysial system. PMID: 19321803
