Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Microtubule-dependent motor required for normal distribution of mitochondria and lysosomes. Can induce formation of neurite-like membrane protrusions in non-neuronal cells in a ZFYVE27-dependent manner. Regulates centrosome and nuclear positioning during mitotic entry. During the G2 phase of the cell cycle in a BICD2-dependent manner, antagonizes dynein function and drives the separation of nuclei and centrosomes. Required for anterograde axonal transportation of MAPK8IP3/JIP3 which is essential for MAPK8IP3/JIP3 function in axon elongation. Through binding with PLEKHM2 and ARL8B, directs lysosome movement toward microtubule plus ends. Involved in NK cell-mediated cytotoxicity. Drives the polarization of cytolytic granules and microtubule-organizing centers (MTOCs) toward the immune synapse between effector NK lymphocytes and target cells.
Gene References into Functions
  1. clathrin promotes formation of phosphatidylinositol(4,5)diphosphate-enriched microdomains, which are required for clustering of KIF5B. PMID: 27219061
  2. These results indicate that KIF5B contributes to mitochondrial localization and function in cardiomyocytes and may play a role in pathological hypertrophic responses in vivo. PMID: 27094714
  3. The authors used an in vitro pulldown assay to define a series of five arginine residues within the conserved pUS9 basic domain of herpes simplex virus 1 that were essential for binding the molecular motor kinesin-1. PMID: 26656703
  4. Diabetes seems to change spinal cord KIF5B and SYD mRNA and sciatic nerves KIF5B content and exercise training modifies it, which may be attributable to the training-induced decreased hyperglycemia. PMID: 25644797
  5. dysferlin-containing vesicles are transported along microtubles via kinesin heavy chain isoform KIF5B, and undergo rapid microtubule- and KIF5B-dependent fusion to form extremely large vesicles following cellular wounding in skeletal muscle cells. PMID: 24203699
  6. Diabetes increased the expression and immunoreactivity of KIF1A and KIF5B in the hippocampus. PMID: 23776493
  7. Therefore, our results reveal that KIF5B is required for the forward transport and axonal function of Nav1.8, suggesting a mechanism for axonal accumulation of Nav1.8 in inflammatory pain. PMID: 24198377
  8. Depletion of kinesin-1 precedes axonal degeneration of the optic nerve in N-methyl-D-aspartate (NMDA)-induced neurotoxicity. PMID: 20863816
  9. Kinesin-12 shares functions with kinesin-5 related to microtubule-microtubule interactions, but kinesin-12 has other functions not shared by kinesin-5 that are related to the ability of kinesin-12 to interact with actin. PMID: 21048148
  10. Binding of JIP1 and FEZ1 to Kinesin-1 is sufficient to activate the motor for microtubule binding and motility. PMID: 17200414
  11. For Kinesin-1, activation requires a global conformational change that separates the kinesin heavy chain (KHC) motor and tail domains and a local conformational change that moves the KHC motor domains closer together. PMID: 17200416
  12. The complex of syntaxin-1-syntabulin-KIF5B mediates axonal transport of the active zone components essential for presynaptic assembly. PMID: 17611281
