To synthesize the recombinant rat purine nucleoside phosphorylase (Pnp) using the baculovirus expression system, the gene fragment encoding the full-length rat Pnp protein (1-289aa) along with a C-terminal 6xHis-tag is cloned into a transfer plasmid. This transfer vector is then co-transfected into insect cells with a baculovirus DNA construct. The co-transfection allows for the homologous recombination of the transfer vector and the baculovirus DNA, resulting in the generation of recombinant baculovirus carrying the rat Pnp gene. Once the recombinant baculovirus is generated, it infects insect cells. The infected cells provide an optimal environment for expressing the recombinant rat Pnp protein. The recombinant rat Pnp protein is isolated and purified from the cell lysate, with up to 90% purity as measured by SDS-PAGE.
