Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Controls the night/day rhythm of melatonin production in the pineal gland. Catalyzes the N-acetylation of serotonin into N-acetylserotonin, the penultimate step in the synthesis of melatonin.
Gene References into Functions
  1. identified one microRNA, miR-325-3p, as a potential candidate responsible for the down regulation of Aanat after survivors of hypoxic-ischemic brain damage PMID: 28502584
  2. these and related results indicate both a major involvement of the N-end rule pathway in the control of rodent AANATs and substantial differences in the regulation of rodent and human AANATs that stem from differences in their N-terminal sequences. PMID: 27339900
  3. Increased methylation and decreased gene expression in the Aanat gene after blast-related TBI in rats. PMID: 25594545
  4. The rhythmic nature of pineal CRX protein may directly modulate the daily profile of Aanat expression by inducing nighttime expression of this enzyme, facilitating nocturnal melatonin synthesis PMID: 24877634
  5. In rats, Aa-nat mRNAs were expressed and existed at high levels during nighttime. They gradually increased at puberty and decreased at 8 weeks of age. Nocturnal changes of Aa-nat mRNA levels in rat pineal gland from P4 to adults were strongly correlated. PMID: 23513468
  6. Local modulation of melatonin synthesis by AANAT may be important for management of the balance between biliary proliferation/damage that is typical of cholangiopathies. PMID: 23080076
  7. miR-483 suppresses Aanat mRNA levels during development and that the developmental decrease in miR-483 abundance promotes melatonin synthesis. PMID: 22908386
  8. The expression of AANAT in epithelial cells of striated ducts in rat salivary glands. PMID: 21437622
  9. Deletion analysis of N-terminus of AANAT identified the an amino acid leucine (L2) as the key residue that contributes to the proteasomal proteolysis of AANAT protein. PMID: 20210853
  10. accumulation induced by pituitary adenylate cyclase activating polypeptide and vasoactive intestinal peptide in vitro PMID: 11854096
  11. inhibition of p38(MAPK), although having no effect on cAMP-mediated AA-NAT transcription, appears to increase AA-NAT activity either by increasing translation or by reducing degradation of the AA-NAT protein. PMID: 14617573
  12. The rapidity of nocturnal AA-NAT induction depends on cAMP inducibility of the gene. Accordingly, cAMP produces a strong AA-NAT response in pineals obtained from rats housed under long photoperiods and a weak AA-NAT response under short photoperiods. PMID: 15046865
  13. aa-nat mRNA increased between 60 and 180 min after the melatonin injection during the light period, while it did not significantly change during the dark period. PMID: 15193530
  14. Effects of protein kinase C on protein stability and activity of rat AANAT were investigated. The results suggest that AANAT was phosphorylated at Thr29 by PKC activation through the alpha(1)-adrenergic receptor in rat pineal glands PMID: 15228600
  15. AANAT mRNA degradation mediated by heterogeneous-muclear ribonucleoproteins hnRNP R, hnRNP Q, and hnRNP L is a key process in the regulation of its circadian oscillation. PMID: 15798208
  16. Important role of proteasomal proteolysis in the adrenergic-mediated induction of aa-nat transcription through its effect on a protein repressor. PMID: 16099857
  17. serotonin N-acetyltransferase regulation by bradykinin in rat pineal glands is affected by adenylyl cyclase VI PMID: 16166080
  18. the effects of the proteasome inhibitors on NE-stimulated Aanat induction appear to be mediated by accumulation of a protein repressor [review] PMID: 16687309
  19. E-box mutations enhanced ectopic expression establishing that the intronic Aanat E-box elements contribute to the gene's pineal specific expression. PMID: 17363136
  20. These findings indicate that the transcriptions of Clock and NAT genes in the pineal gland not only show remarkably synchronous endogenous circadian rhythmic changes, but also respond to the ambient light signal in a reduced manner. PMID: 17364576
