Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Component of the SUMO-targeted ubiquitin ligase (STUbL) complex SLX5/SLX8 that mediates ubiquitination and subsequent desumoylation of sumoylated proteins and proteins containing SUMO-like domains for their degradation. The STUbL complex SLX5/SLX8 stimulates ubiquitin conjugating enzymes, including UBC1, UBC4, UBC5 and UBC13-MMS2, and mediates the proteolytic down-regulation of sumoylated proteins. The STUbL complex SLX5/SLX8 is involved in ubiquitin-mediated degradation of histone variant CSE4, preventing mislocalization to euchromatin. The complex plays an essential role in maintenance of chromosome stability and links SUMO-dependent ubiquitination to a centromere-specific function during mitosis. The complex is involved in proteolysis of spindle positioning protein KAR9 and ensures correct spindle function by regulating levels of microtubule-associated proteins. During replication, the complex helps prevent DNA lesions via recombination and has a role in localizing the DNA damage protein DCD2. The complex especially ubiquitinates the nuclease YEN1 and prevents persistent accumulation of a fraction of YEN1 associated with sites of activity in late G2/M and helps maintain the balance between pro- and anti-crossover pathways during homologous recombination. It is also involved in ubiquitin-mediated degradation of DNA repair proteins RAD52 and RAD57. Along with SIR2, promotes silencing of genes at telomeric or ribosomal DNA (rDNA) loci. Finally, the complex is recruited to distinct genomic hotspots of non-H2B protein ubiquitination (ub-hotspots) by the sumoylated transcription factor-like protein EUC1 where it ubiquitinates EUC1 and presumably other targets.
Gene References into Functions
  1. SLX5 suppressor mutations of tdp1 wss1 synthetic lethality created new lysine residues in a 398 amino acid domain that is completely free of lysine. They result in the auto-ubiquitination and partial proteolysis of Slx5. PMID: 28550017
  2. RecQ-like helicase Sgs1 subcellular localization is regulated by SUMO-targeted ubiquitin ligase Slx5-Slx8 in response to DNA damage, presumably to prevent illegitimate recombination events. PMID: 25588990
  3. Siz1 can be ubiquitylated in vivo and is degraded in an Slx5-dependent manner. PMID: 24196836
  4. distinct SUMO ligases work in concert with Esc2 and Slx5-Slx8 to control substrate specificity and sumoylation homeostasis to prevent GCRs. PMID: 23935535
  5. Component or regulator of the small ubiquitin-related modifier (SUMO) protein pathway. PMID: 16387868
  6. Data suggest that, during replication, the Slx5-Slx8 complex helps prevent DNA lesions that are acted upon by recombination, and inhibits Rad51-independent recombination via modulating the sumoylation of DNA repair proteins. PMID: 17591698
  7. the Slx5 and slx8 complex is capable of interacting directly with the SUMO pathway PMID: 17669696
  8. the Hex3.Slx8 complex has a robust substrate-specific E3 ubiquitin ligase activity. PMID: 17848550
  9. The Slx5-Slx8 complex, but not its individual units, stimulates several ubiquitin ligase conjugating enzymes. PMID: 18032921
  10. Slx5-Slx8 ubiquitin ligase is stimulated by poly-SUMO conjugates and that it can ubiquitinate a poly-small ubiquitin-like modifier chain PMID: 18499666
  11. study shows a nuclear pore subcomplex and Slx5/Slx8, a SUMO-dependent ubiquitin ligase physically interact; findings suggest that DNA strand breaks are shunted to nuclear pores for a repair pathway controlled by a conserved SUMO-dependent E3 ligase PMID: 18948542
  12. the DNA-damage targeting of Slx5/Slx8 depends on formation of the heterodimer and that this occurs at a subset of nuclear foci also containing DNA damage repair and checkpoint factors. PMID: 19270524
