Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Master transcriptional regulator that mediates the response to amino acid starvation. Binds variations of the DNA sequence 5'-ATGA[CG]TCAT-3' in canonical nucleosome-depleted 5'-positioned promoters, and also within coding sequences and 3' non-coding regions. During nutrient starvation (low or poor amino acid, carbon or purine sources), it activates genes required for amino acid biosynthesis and transport, autophagy, cofactor biosynthesis and transport, mitochondrial transport, and additional downstream transcription factors. Activates transcription by recruiting multiple coactivators, including the mediator complex, the SAGA complex, and the SWI/SNF complex, to enable assembly of the pre-initiation complex at core promoters.
Gene References into Functions
  1. The down-regulation of the GCN4 expression (Gcn(-) phenotype) in the eIF5(G31R) mutant was not because of leaky scanning defects; rather was due to the utilization of upUUG initiation codons at the 5' regulatory region present between uORF1 and the main GCN4 ORF. PMID: 28385532
  2. GCN4 is required for nitrogen catabolite repression-sensitive nuclear Gln3 localization. PMID: 28007891
  3. Gcn4 sumoylation in limiting its association with chromatin. PMID: 27770033
  4. Presented are molecular dynamics simulations reflecting a secondary structure of phopspho-GCN4 at different pH. PMID: 28230348
  5. Reinitiation on GCN4 mRNA after translation of its four short uORFs is regulated by the sequence immediately following their respective stop codons. PMID: 26822200
  6. Upstream-open reading frame 2 (uORF2) from the GCN4 mRNA leader resembles the well-established reinitiation (REI)-permissive uORF1 in its high REI-promoting activity. PMID: 24623812
  7. our results suggest that the folding behavior of the well-studied GCN4 leucine-zipper domain is more complex than was previously appreciated. PMID: 23116373
  8. Elucidated RNA binding interface of the GCN4 leucine zipper motif from yeast. Site-directed mutagenesis of the LZ-GCN4 RNA binding interface showed that substrate binding is facilitated by lysine and arginine side chains. PMID: 22374868
  9. the combination of Gln3 and Gcn4 results in the constitution of a hybrid modulator which elicits a novel transcriptional response, not evoked when these modulators act in a non-combinatorial fashion. PMID: 21184740
  10. Gcn4(L267S) variant carries an amino acid substitution in the third of four conserved leucines of the zipper dimerization domain. This abolished FLO11 expression and resulted in reduced transcriptional activity for amino acid biosynthetic genes. PMID: 21111745
  11. GAAC is a major effector of the TOR pathway, with Gcn4p and Gln3p each inducing a similar number of genes during rapamycin treatment PMID: 20233714
  12. interactions between the two nanorope-like assemblies are due to the hydrophobic residues on the leucine faces, which are also responsible for the dimer formation of the wild-type GCN4 leucine zipper. PMID: 20027625
  13. eIF3b functions between 48S assembly and subunit joining to influence AUG recognition and reinitiation on GCN4 mRNA PMID: 14976554

  19. large fraction of genes activated by Gcn4p are repressed by H3 and H4 amino termini under non-inducing conditions, indicating that one function of Gcn4p is to overcome chromatin-mediated repression that depends on histone tails PMID: 16461773
  20. report here the design of a modified GCN4-bZIP DNA-binding protein with an azobenzene chromophore introduced between Cys residues at positions 262 and 269 (S262C, N269C) within the zipper domain PMID: 16681380
