Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Histone chaperone that facilitates histone deposition and histone exchange and removal during nucleosome assembly and disassembly. Facilitates histone deposition through both replication-dependent and replication-independent chromatin assembly pathways. Cooperates with chromatin assembly factor 1 (CAF-1) to promote replication-dependent chromatin assembly and with the HIR complex to promote replication-independent chromatin assembly, which may occur during transcription and DNA repair. May be required for the maintenance of a subset of replication elongation factors, including DNA polymerase epsilon, the RFC complex and PCNA, at stalled replication forks. Also required for RTT109-dependent acetylation of histone H3 on 'Lys-9' and 'Lys-56'. Promotion of RTT109-mediated histone H3 'Lys-56' acetylation is dependent on interactions with histone H3 pre-acetylated on 'Lys-14'.
Gene References into Functions
  1. he model provides a molecular basis for the involvement of both Vps75 and Asf1 in Rtt109 catalysed histone H3 K9 acetylation. In the absence of Asf1 this model can be used to generate a complex consisting of a reconfigured Vps75 tetramer bound to a H3-H4 tetramer. PMID: 27036862
  2. dissociation of histone H3 from Asf1 is required for efficient recovery and that Asf1 is required for complete dephosphorylation of Rad53 when the upstream DNA damage checkpoint signaling is turned off PMID: 27222517
  3. These results along with high association of Asf1 with heterochromatic centromeres and telomeres ascribe novel roles to Asf1 in condensin loading and chromatin dynamics. PMID: 25264624
  4. acetylation of histone H4 K12 by Rtt109-Vps75, whereas Rtt109-Asf1 showed no detectable activity toward H4. PMID: 23036725
  5. A region (amino acids [aa] 188 to 231) within the C terminus of Asf1 is required for its normal function in transcriptional silencing. PMID: 23184661
  6. Asf1 and Snf2 function to move kinetically trapped nucleosomes to their energetically favorable sites in promoters PMID: 21508315
  7. Saccharomyces cerevisiae Asf1 shields H3/H4 from unfavorable DNA interactions and aids the formation of favorable histone-DNA interactions through the formation of disomes. PMID: 21447559
  8. Asf1 stimulates derepression of DNA damage response (DDR) genes during the S phase PMID: 21190944
  9. Results suggest that defective chromatin assembly caused by Asf1 leads to double-strand breaks that can be repaired by sister chromatid exchange, affecting genetic stability. PMID: 15071494
  10. may have a global role in chromatin disassembly and an unexpected role in histone acetylation in vivo PMID: 15452122
  11. ASF1 has a role in activation of the DNA damage checkpoint PMID: 15542840
  12. Data indicate that the histone chaperones CAF-1 and Asf1p regulate the gene expression of a broad array of genes in yeast and, in the case of Asf1p, this is likely to be due to a direct role in chromatin modulation during transcriptional regulation. PMID: 15632066
  13. ASF1 is required for the recruitment of the SAS histone acetyltransferase complex to the HMLalpha locus. PMID: 15891116
  14. Restoration of silencing to CAF-1 subunit cac1 Delta cells requires both Asf1 and Hir1 PMID: 16020781
  15. Asf1 blocks formation of the H3-H4 heterotetramer by a mechanism that likely involves occlusion of the H3-H3 dimerization interface PMID: 16229457
  16. HIR complex and Asf1 proteins function together as a conserved eukaryotic pathway for histone replacement throughout the cell cycle. PMID: 16303565
  17. Together, these data indicate that CAF-1 interacts with H3 methylated at lysine 79 during the processes of epigenetic silencing and DNA repair. PMID: 16503640
  18. identified mutations in the nonconserved C terminus of antisilencing function 1 (Asf1) that result in enhanced silencing of HMR and telomere-proximal reporters, overcoming the requirement for CAF-1 in transcriptional silencing PMID: 16582440
  19. These data suggest that maintenance of histone H3 lysine 56 acetylation is a primary contribution of Asf1 to genome stability in yeast. PMID: 16627621
  20. These results strongly suggest that Asf1 functions as an elongation factor to disassemble and reassemble histones during Pol II elongation. PMID: 16678113
  21. Asf1 does not affect the stability of the newly synthesized histones per se, but instead histone binding by Asf1 promotes the efficient acetylation of specific residues of newly synthesized histone H3 PMID: 17107956
  22. Asf1 contributes to interpretation of a "histone H3 barcode" for sorting H3 isoforms into different deposition pathways. PMID: 17166288
  23. CAF-1 and Asf1 function redundantly to deactivate the checkpoint by restoring chromatin structure on the completion of DNA double-strand break repair. PMID: 19164567
  24. Results suggest that the SWI/SNF complex is directly involved in nucleosome removal at HO, and that the histone chaperone Asf1 assists in this during the course of gene regulation. PMID: 19470759
  25. ASF1 probably contributes to the maintenance of genome stability through multiple mechanisms, some of which involve the postreplication repair and homologous recombination suppression pathways PMID: 19635810
  26. The histone chaperone Rtt106 functions in a pathway with two other chaperones, Asf1 and the HIR complex, to create a repressive chromatin structure at core histone promoters. PMID: 19683497
