Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Probably involved in mitochondrial protein import. Is also required for efficient translocation of pre-pro-alpha-factor. Involved in heme regulation of HAP1, as a component of the high-molecular-weight (HMC) complex.
Gene References into Functions
  1. The authors provide genetic, biochemical, and biophysical evidence that the Ydj1p CaaX motif is isoprenylated but not cleaved and carboxylmethylated. PMID: 27525482
  2. Ydj1 is required for the formation of Dhh1 complexes. PMID: 26392412
  3. Ydj1 directs disaggregation activity of the Hsp100-based machinery to provide stress protection. PMID: 25329162
  4. The chaperone Ydj1 interacts specifically with mutant septins during septin quality control. PMID: 25673805
  5. Results show that Hsp70 and the Ydj1 and Sis1 Hsp40 proteins accumulated in stress granules and defects in these proteins led to decreases in the disassembly and/or clearance of stress granules. PMID: 26199455
  6. Ydj1 is a yeast Type I Hsp40 in a highly conserved alpha-helical N-terminal J-domain, a disordered middle region (a glycine/phenylalanine (G/F)), a Zinc finger domain (ZFLR or Cys-rich domain) and a two C-terminal sub-domains. PMID: 22011374
  7. Ydj1p/Hlj1p catalyzes CFTR degradation PMID: 15342786
  8. the hydrophobic pocket located on Ydj1 plays a critical role in its molecular chaperone activity by mediating interactions with the non-native polypeptides PMID: 15500443
  9. crystal structure of the C-terminal fragment of yeast Hsp40 Ydj1 that is responsible for Ydj1 dimerization PMID: 15701512
  10. The introduction of YDJ1p improves secretory proteins in Pichia pastoris secretion 4-7 times. PMID: 16889384
  11. ydj1delta yeast exhibit phenotypes consistent with cell-wall defects, and these phenotypes were improved by Mid2p or Pkc1p overexpression PMID: 17237519
  12. Genetic experiments showed that Ydj1 is not essential for viability; however, Stability experiments revealed that only Ydj1-protected Hsp70 proteins can hydrolyze ATP under prolonged stress. PMID: 17985367
  13. Ydj1 is identified as being important for maintaining steady-state levels of protein kinases after screening several chaperones and cochaperones in gene deletion mutant strains. PMID: 18443039
  14. Results indicate that farnesylation is a major factor contributing to the specific requirement for Ydj1 in promoting proper regulation and activation of diverse Hsp90 clients. PMID: 18829866
  15. The combined action of a farnesyl moiety and zinc finger-like region enable Type I Hsp40s to recognize amyloid-like substrates and prevent formation of cytotoxic protein species. PMID: 19056735
  16. A unique motif that is selectively recognized by Ydj1p, was computationally identified. PMID: 19549854
