  • IHC image of CSB-RA035034A0HU diluted at 1:100 and staining in paraffin-embedded human lung cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB.

  • Overlay Peak curve showing jurkat cells stained with CSB-RA035034A0HU (red line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100 for 10min. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 35min at 4℃.Control antibody (green line) was Rabbit IgG (1ug/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.

Description

ANP32B, also known as acidic leucine-rich nuclear phosphoprotein 32 family member B, plays important roles in chromatin remodeling, cell proliferation, and apoptotic regulation. This multifunctional protein has garnered significant research interest due to its involvement in transcriptional regulation and its emerging connections to cancer biology, making it a valuable target for investigators studying cellular homeostasis and disease mechanisms.

This recombinant monoclonal antibody, clone 8C9, offers the consistency and reliability that demanding experimental workflows require. Produced using recombinant technology with a defined sequence, this antibody eliminates the lot-to-lot variability that can compromise longitudinal studies and reproducibility. The rabbit host-derived IgG format provides excellent signal-to-noise characteristics while maintaining compatibility with standard secondary detection systems.

Validation data demonstrates robust performance across multiple applications. Western blot analysis confirms specific detection of ANP32B at the expected 29 kDa molecular weight across an extensive panel of human cell lines, including HT-29, THP-1, U-251MG, A549, K562, MCF-7, Jurkat, A431, and HeLa cells, providing confidence when working with diverse cellular models. Immunohistochemistry validation in paraffin-embedded human lung cancer and testis tissues shows reliable tissue staining with citrate buffer antigen retrieval, while flow cytometry analysis in Jurkat cells demonstrates clear population separation from isotype controls, confirming utility for single-cell analysis applications.

The antibody is supplied in a glycerol-containing buffer optimized for long-term storage stability. Whether investigating ANP32B's role in chromatin dynamics, exploring its function in proliferative signaling, or examining its expression patterns in tumor biology, this antibody provides the technical foundation for meaningful experimental insights.

Target Background

Function
Multifunctional protein that is involved in the regulation of many processes including cell proliferation, apoptosis, cell cycle progression or transcription. Regulates the proliferation of neuronal stem cells, differentiation of leukemic cells and progression from G1 to S phase of the cell cycle. As negative regulator of caspase-3-dependent apoptosis, may act as an antagonist of ANP32A in regulating tissue homeostasis. Exhibits histone chaperone properties, able to recruit histones to certain promoters, thus regulating the transcription of specific genes. Plays also an essential role in the nucleocytoplasmic transport of specific mRNAs via the uncommon nuclear mRNA export receptor XPO1/CRM1. Participates in the regulation of adequate adaptive immune responses by acting on mRNA expression and cell proliferation.; (Microbial infection) Plays an essential role in influenza A and B viral genome replication. Plays also a role in foamy virus mRNA export from the nucleus to the cytoplasm.
