  • IHC image of CSB-RA207790A0HU diluted at 1:100 and staining in paraffin-embedded human liver cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB.

Description

The production of the STK33 recombinant monoclonal antibody follows a stringent and well-defined process to ensure its quality and specificity. Initially, B cells are isolated from an immunized animal, with the synthesized peptide derived from human STK33 used as the immunogen. Subsequently, total RNA is extracted from the isolated B cells and converted into cDNA through reverse transcription. The STK33 antibody genes are then amplified using PCR with specific primers targeting the antibody constant regions and inserted into an expression vector. This vector is introduced into host cells, facilitating the production of the STK33 recombinant monoclonal antibody. The antibody is harvested from the cell culture supernatant and purified using affinity chromatography, resulting in a highly purified form. Rigorous characterization assays, including ELISA and IHC analysis, are performed to validate the antibody's specificity and functionality, ensuring its precise recognition of human STK33 protein.

Target Background

Function
Serine/threonine protein kinase which phosphorylates VIME. May play a specific role in the dynamic behavior of the intermediate filament cytoskeleton by phosphorylation of VIME. Not essential for the survival of KRAS-dependent AML cell lines.
Gene References into Functions
  1. Dysregulated HIF1alpha/STK33 signaling promotes pancreatic ductal adenocarcinoma (PDAC) growth and progression, this suggests that STK33 is a candidate therapeutic target to improve PDAC treatment. PMID: 29038348
  2. STK33 is a potential oncogene and plays an important role in tumorigenesis of HSCC via regulation of numerous genes. PMID: 27414193
  3. STK33 plays an essential role in hepatocellular proliferation and liver tumorigenesis. STK33 may bind directly to c-Myc and increase its transcriptional activity. PMID: 25398772
  4. Results show that STK33 is a potential oncogene and a promising diagnostic marker for hypopharyngeal squamous cell carcinoma (HSCC). STK33 may promote tumorigenesis and progression of HSCC, and serve as a valuable molecular target for treatment of HSCC. PMID: 25603720
  5. STK33 can promote cell migration and invasion and suppress p53 gene expression in the NL9980 and L9981 large cell lung cancer cell lines. In addition, this protein also promotes epithelial-mesenchymal transition. PMID: 25662617
  6. The STK33-linked SNP rs4929949 is associated with obesity and BMI in two independent cohorts of Swedish and Greek children. PMID: 23967198
  7. STK33 downregulation or dominant mutant overexpression had no effect on KRAS signaling or survival of cancer cells. PMID: 21742770
  8. STK33/Stk33 expression pattern resembles those of some related members of the calcium/calmodulin dependent kinase group PMID: 16176263
