  • IHC image of CSB-RA903925A0HU diluted at 1:100 and staining in paraffin-embedded human melanoma cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB.

  • Overlay Peak curve showing SH-SY5Y cells stained with CSB-RA903925A0HU (red line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100 for 10min. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 35min at 4℃.Control antibody (green line) was Rabbit IgG (1ug/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.

Description

Tropomodulin-2, also known as neuronal tropomodulin, plays a critical role in regulating actin filament dynamics by capping the pointed ends of actin filaments in neurons. This function is essential for maintaining neuronal architecture, synaptic plasticity, and proper nervous system development. Researchers investigating neurodegenerative diseases, neuronal cytoskeletal organization, and brain development will find TMOD2 a compelling target for understanding how actin dynamics influence neuronal function and pathology.

This recombinant monoclonal antibody, generated from clone 10B12, offers the reproducibility and consistency that demanding research protocols require. Because the antibody sequence is defined and production occurs in controlled recombinant systems, you can expect reliable performance across experiments and over time, eliminating the lot-to-lot variability that can complicate long-term studies or multi-site collaborations.

Validation studies demonstrate this antibody's versatility across multiple experimental platforms. Immunohistochemistry performed on paraffin-embedded human tissues reveals clear staining in both brain tissue, where TMOD2 is abundantly expressed, and melanoma samples, suggesting potential utility in cancer research contexts. Flow cytometry analysis using the SH-SY5Y neuroblastoma cell line, a widely used model for neuronal studies, shows distinct positive signal separation from isotype controls, confirming specific intracellular detection following fixation and permeabilization.

The antibody is supplied in a glycerol-containing buffer optimized for long-term stability at freezer temperatures, and affinity purification ensures high purity for sensitive downstream applications. Whether you are mapping TMOD2 expression patterns in clinical specimens, characterizing neuronal differentiation in cell culture models, or exploring its involvement in disease states, this antibody provides a dependable tool for advancing your research into actin-based neuronal biology.

Target Background

Function
Blocks the elongation and depolymerization of the actin filaments at the pointed end. The Tmod/TM complex contributes to the formation of the short actin protofilament, which in turn defines the geometry of the membrane skeleton.
Subcellular Location
Cytoplasm, cytoskeleton.
Protein Families
Tropomodulin family
Tissue Specificity
Neuronal-tissue specific.
