In the in vitro E. coli expression system, the recombinant rat sphingosine 1-phosphate receptor 2 (S1pr2) is produced through the gene fragment that encodes the full-length rat S1pr2 protein (1-352aa). The process involves cloning the S1pr2 gene fragment along with a C-terminal 10xHis-tag into a suitable expression vector with the necessary regulatory elements. The recombinant plasmid DNA is then isolated and purified. In vitro transcription is performed using the purified plasmid DNA to generate mRNA, which serves as a template in the subsequent in vitro translation reaction. The translation reaction is carried out using an E. coli cell-free extract supplemented with energy sources and amino acids. After incubation, the recombinant S1pr2 protein is purified from the cell culture medium. The purified protein exhibits a purity level of up to 85% as confirmed by SDS-PAGE analysis. On the gel, the recombinant S1pr2 protein appears as a distinct band with a molecular weight of approximately 35 kDa.
