We are solving topology of a membrane protein (5TM) now and want to know whether you have the capacity to express the protein. A pilot test will be performed first for an evaluation. We prefer to use baculovirus system in sf9 cells. Function is similar in yeast, human and all higher eukaryotes.

Attached is the sequence of the plasmid in the area of the Surf4 insert
Yeast sequence of homologous protein, ERV29, is P53337.1
We have fully functional Surf4 when we add a HA tag immediately after the starting methionine. I’d be hesitant to put anything longer there as this is one of those proteins where the amino terminus moves into the lumen of the ER without a leader sequence.
Unknown if functions as dimer or higher order but is found in dimers, trimers etc on western blots unless overnight incubation in LDS and 8M urea is used. We have binding assay but works to date only in native membrane preps, no binding yet when fully solubilized with n-Decyl-β-D-Maltopyranoside. May be in low affinity form when in DM or may not be folded correctly at all in this detergent.
Detailed plan is not necessary at this time, just a ball-park costs of various logical steps in your process. I have cDNA for human and yeast. If possible, could you please provide me an estimated cost and time for a pilot expression test of either/both of the proteins.
