Target Background

Function
Acts as a sensor that couples lifespan to information about energy levels and insulin-like signals. Role in motility and response to oxidative stress. Involved in the establishment of germline stem cell (GSC) quiescence during dauer development. Plays a role in axon regrowth after axotomy in PLM neurons. Plays a role in the maintenance of glycogen stores which are necessary for resistance to hyperosmotic stress. Plays a role in the regulation of flp-7 secretion from ASI neurons. Keeps the CREB-regulated transcription coactivator 1 homolog crtc-1 inactive which in turn inhibits flp-7 secretion. Following serotonin signaling, derepresses crtc-1 which stimulates flp-7 secretion and subsequent body fat loss.
Gene References into Functions
  1. Knockdown of Indy/CeNAC2 increased life span by 22+/-3 % compared to controls, together with a decrease in whole body fat content by ~50%. Indy/CeNAC2 reduction also increased the activation of the intracellular energy sensor AMPK/aak2. PMID: 26318988
  2. As in elevated serotonin signaling, inactivation of AAK-2 in the ASI neurons caused enhanced secretion of dense core vesicles from these neurons. PMID: 24921650
  3. An essential output of RSKS-1 in axon regrowth is the metabolic sensor AMP kinase, AAK-2. PMID: 24431434
  4. AMPK catalytic subunit aak-2 mutants exhibited elevated pharyngeal pumping even in the absence of exogenous 5-HT. PMID: 22768843
  5. The capacity for an animal to survive long bouts of severe oxygen deprivation is likely dependent on specific subunits of the heterotrimeric protein AMPK and energy stores such as carbohydrates. PMID: 21304820
  6. These results show that aging is controlled by overlapping but distinct pathways and that AMPK/aak-2 represents a node in a network of evolutionarily conserved biochemical pathways that control aging. PMID: 16626391
  7. C. elegans larvae that lack LKB1/AMPK (AMP-activated protein kinase) signalling enter dauer normally, but then rapidly consume their stored energy and prematurely expire following vital organ failure PMID: 19052547
  8. aak-2 is required for life span extension in states of impaired glycolysis and calorie restriction. aak-2 initiates increased mitochondrial respiration and increased oxidative stress/ROS formation to promote mitohormesis. PMID: 17908557
