Target Background

Function
Part of a checkpoint which monitors spindle integrity and prevents premature exit from mitosis. This cell-cycle arrest depends upon inhibition of the G-protein TEM1 by the BFA1/BUB2 complex.
Gene References into Functions
  1. Tem1 activity is tightly controlled in mitosis by Bub2 and Bfa1 and is also regulated by the spindle orientation checkpoint that monitors the correct alignment of the mitotic spindle with the mother-daughter axis. Here is described the purification of Tem1, Bfa1, and Bub2 and a detailed radioactive filter-binding assay to study the nucleotide binding properties of Tem1 and the role of its regulators Bfa1 and Bub2. PMID: 27826857
  2. Bmh1 is a spindle position checkpoint (SPOC) component that breaks Bfa1 centrosome asymmetry upon SPOC activation. PMID: 24850890
  3. Cdc5 induces the asymmetric distribution of Bfa1 to the bud-directed spindle pole body independently of Bfa1 GAP activity at anaphase. PMID: 22253605
  4. various checkpoint signals are transmitted to the C-terminal domain of Bfa1 (Bfa1-D8(391-574)) and Bfa1p localization to spindle pole body is necessary but not sufficient to regulate mitotic exit in response to various checkpoint signals PMID: 15147270
  5. Bfa1 disappearance from the mother bound spindle pole body at the onset of anaphase, along with Cdc5 function, helps prompt Tem1 activation in telophase. PMID: 16449187
  6. Results suggest that the GTPase-activating activity of Bfa1p-Bub2p is responsible for the mitotic arrest caused by spindle damage and Bfa1p overproduction. PMID: 16682821
  7. there is a GAP-independent surveillance mechanism of Bfa1/Bub2, which, together with the GTP/GDP switch of Tem1, may be required for the genomic stability of cells with misaligned spindles PMID: 18667533
  8. A link is confirmed between the subcellular localization pattern of Bfa1 at the spindle pole bodies with cell cycle progression. PMID: 19948498
