Target Background

Function
Activator protein that regulates the ubiquitin ligase activity and substrate specificity of the anaphase promoting complex/cyclosome (APC/C). At the metaphase-to-anaphase transition, recognizes and binds proteins containing a D-box including the B-type cyclins CLB2 and CLB5, HSL1 and securin PDS1, and recruits them in a C-box-dependent manner to the APC/C for ubiquitination and subsequent proteolysis. Required for sister chromatid separation and disassembly of the mitotic spindle. Target of the spindle checkpoint pathway through participation in the mitotic checkpoint complex (MCC) and the MAD2-CDC20 subcomplex. MCC and presumably the MAD2-CDC20 subcomplex inhibit the ubiquitin ligase activity of the anaphase promoting complex/cyclosome (APC/C) by preventing its activation by CDC20 until proper attachment of all chromosomes to the spindle.
Gene References into Functions
  1. Small N-terminal regions of both Cdc20 (RxxL-type D box) and Cdc5 (KEN motif) were identified as transferable degradation domains (degrons) that mediated efficient targeting by APC/C-Cdh1 (anaphase-promoting complex/cyclosome-Cdh1). PMID: 25540434
  2. Bub3 has a previously unknown function at the kinetochore in activating APC/C-Cdc20 for normal mitotic progression. PMID: 25987604
  3. Both catalysis of Mad1:C-Mad2 at the kinetochores and Mad2-Cdc20 binding away from kinetochores are required for a proficient checkpoint. PMID: 23000150
  4. study analyzed the role of Cdc20 autoubiquitination in the spindle assembly checkpoint PMID: 22940250
  5. This study finds that the majority of Cdc20 turnover does not involve a second activator molecule but instead depends on in cis Cdc20 autoubiquitination while it is bound to its activator-binding site on the anaphase-promoting complex (APC) core. PMID: 22079111
  6. Mad3 is required for the stable binding of Mad2 to Cdc20 in vivo, which is sufficient to inhibit APC activity and is the most downstream event in spindle checkpoint activation. PMID: 22209528
  7. data suggest important roles of ubiquitination in mediating IGF-1R signaling and degradation PMID: 17406666
  8. model aimed at unifying previous analyses of checkpoint function by focusing on the Mad3-Cdc20 interaction PMID: 17369399
