Target Background

Function
Component of the ribosomal small subunit processome for the biogenesis of ribosomes, functions in pre-ribosomal RNA (pre-rRNA) processing. Essential for embryonic development in part through the regulation of p53 pathway. Required for the expansion growth of intestine, liver and exocrine pancreas, but not endocrine pancreas. May not play a major role during the early stage of endodermal organogenesis. Also involved in the sympathetic neuronal development. Triggers the degradation of p53/TP53 and Delta113p53 isoform in a ubiquitination pathway independent way. Mediates, with CAPN3, the proteasome-independent degradation of p53/TP53.
Gene References into Functions
  1. Fibrotic scar formation in def+/- is blocked by the over-expression of Def. PMID: 24801718
  2. Def can mediate the degradation of p53 protein and that this process is independent of the proteasome pathway, but dependent on the activity of Calpain3, a cysteine protease. PMID: 23357851
  3. Def functions in a cell autonomous manner during early liver development. PMID: 23593122
  4. def gene encodes a novel pan-endoderm-specific factor.p53 is tightly regulated by factor Def, especially during organogenesis PMID: 16322560
Subcellular Location
Nucleus, nucleolus.
Protein Families
Def family
Tissue Specificity
Mainly expressed in digestive organs at the later stage of organogenesis. Expressed in liver, gut and exocrine pancreas, but not in the islets.
