Target Background

Function
In addition to polymerase activity, this DNA polymerase exhibits 3'-5' and 5'-3' exonuclease activity. It is able to utilize nicked circular duplex DNA as a template and can unwind the parental DNA strand from its template.
Gene References into Functions
  1. Findings indicate an unrealized function of DNA polymerase I (Pol I), and a possible mechanism by which G4 structures can be resolved during DNA replication and/or repair. PMID: 28986969
  2. Pol I catalyzes DNA synthesis past N6-dA-linked peptide cross-links and is likely to play an role in cellular bypass of large major groove DNA lesions. PMID: 21622737
  3. analysis of complexes of DNA polymerase I (Klenow fragment) for base and sugar discrimination during nucleotide substrate selection PMID: 21362617
  4. a new in vivo function for wild-type DNA polymerase I in chromosome termination at site(s) not yet identified is proposed PMID: 15752206
  5. mechanism in which Arg668 forms a hydrogen bonding fork between the minor groove of the primer terminus and the ring oxygen of the deoxyribose moiety of the incoming dNTP to align the 3'-hydroxyl group with the alpha-phosphate of the dNTP PMID: 15823023
  6. Glutamine677 is perhaps the first residue in the pol I family of enzymes that is shown to be essential for the functioning of both polymerase and proofreading activities. PMID: 15924429
  7. mutant(Delta)polA is defective in base mismatch correction PMID: 16005896
  8. analysis of amino acid substitutions that increase the fidelity of Escherichia coli DNA polymerase I PMID: 17301051
  9. Data show that dnaX36 strains have an increased requirement for DNA polymerase I. PMID: 18156258
