Target Background

Function
Essential signaling protein which has a major role in germline and embryonic development; involved in cell fate decisions that require cell-cell interactions. Probable membrane-bound receptor for putative ligands lag-2 and apx-1. Upon ligand activation, and releasing from the cell membrane, the glp-1/Notch intracellular domain (NICD) probably forms a transcriptional activator complex with lag-1 and lag-3 and regulates expression of various genes; targets in the germline include lst-1 and sygl-1. Involved in the specification of the cell fates of the blastomeres, ABa and ABp. Proper signaling by glp-1 induces ABa descendants to produce anterior pharyngeal cells, and ABp descendants to adopt a different fate. Contributes to the establishment of the dorsal-ventral axis in early embryos. Required in postmitotic neurons in order to maintain the developmentally arrested larval state known as dauer, probably in response to lag-2. Regulates germ cell mitotic proliferation probably by regulating MAP kinase phosphatase lip-1 expression. Required for oocyte growth control. Plays a negative role in lifespan.
Gene References into Functions
  1. These results provide the first experimental evidence of an essential role for HSP90 in Notch signaling in development. PMID: 29507057
  2. The authors find that Notch-dependent active transcription sites occur in a probabilistic fashion and, unexpectedly, do so in a steep gradient across the stem cell pool. PMID: 27705743
  3. Zygotically expressed MOM-2 (Wnt) is responsible for two distinct cell-cell interactions as the MS blastomere produces one-third of the body wall muscles. Both the inhibitory and the activating interactions require precise spatiotemporal expression of zygotic MOM-2, which is dependent upon two distinct GLP-1 Notch signals. PMID: 28049659
  4. Study suggests that GLP-1 signaling is directly responsible for driving all stages of proliferation in the wild-type germline, and loss of its activity causes differentiation without intervening, distinctly regulated transit-amplifying divisions. PMID: 26158953
  5. glp-1(ar202), a temperature-sensitive gain-of-function C. elegans NOTCH mutant, was employed to delineate NOTCH-driven tumor responses to radiotherapy. PMID: 26120834
  6. regulatory element is highly conserved in the paralogous glp-1 genomic environment, suggesting redundant developmental and physiological roles for the two Notch paralogs in the C. elegans nervous system. PMID: 23305862
  7. POS-1 regulates glp-1 mRNA translation by blocking access of other RNA-binding proteins to a key regulatory sequence PMID: 23034181
  8. The functional defect in glp-1(Pro) mutants occurs prior to or at the level of ligand interaction. PMID: 12586701
  9. the balance between POS-1 and SPN-4 controls the translation of maternal glp-1 mRNA PMID: 12702662
  10. Results suggest that TBX-37 and TBX-38 play a key role in distinguishing the outcomes of two sequential Glp-1/Notch-mediated interactions. PMID: 15056620
  11. Data support a model in which GLP-1 function in MSP-dependent oocyte growth is separable from its role in the proliferation versus meiotic entry decision. PMID: 19502484
