Description

The preparation of polyclonal antibodies targeting HBZ involves the iterative immunization of a rabbit with recombinant human T-cell leukemia virus 1 (isolate Caribbea HS-35 subtype A) (HTLV-1) HBZ protein (1-209aa). Following the attainment of an adequate antibody titer, the rabbit is bled, and antibodies are purified from the serum utilizing affinity chromatography. The efficacy of the resulting HBZ antibody is then assessed in ELISA and WB applications, revealing its reactivity with HTLV-1 (isolate Caribbea HS-35 subtype A) HBZ protein.

The HTLV-1 HBZ protein is a pivotal regulator in the HTLV-1 life cycle, influencing the pathogenesis of HTLV-1-associated diseases. Primarily functioning as a transcriptional regulator, HBZ modulates the expression of viral and host genes, contributing to the altered gene profile observed in HTLV-1-infected cells. Beyond its role in transcriptional regulation, HBZ is implicated in viral replication, maintaining a balance between viral reproduction and host immune responses for prolonged viral persistence. Additionally, HBZ is crucial for immune evasion, influencing T cell function and other immune responses to facilitate chronic infection.

Target Background

Function
Enhances viral infectivity and persistence, and facilitates proliferation of HTLV-1-infected lymphocytes. Mechanistically, inhibits Tax-mediated viral replication and NF-kappa-B activation. Plays a role in allowing infected T-cells to escape the cytotoxic T-lymphocyte response by maintaining low levels of viral protein production. Inhibits also host EP300 histone acetyltransferase (HAT) activity, reducing levels of acetylated histone H3 at 'Lys-18' (H3K18ac) in infected cells. Contributes to the accumulation of chromosomal abnormalities by inhibiting double-stranded DNA breaks (DSB) repair through the NHEJ pathway. Participates in the modulation of host immune response at multiple levels contributing to abnormal interferon signaling and viral pathogenesis.
