Target Background

Function
Importin beta subunit that functions in nuclear protein import through association with the importin alpha subunit, which binds to the classical nuclear localization signal (cNLS) in cargo substrates. Docking of the importin/substrate complex to the nuclear pore complex (NPC) is mediated by importin beta through binding to nucleoporin FxFG repeats and the complex is subsequently translocated through the pore by an energy requiring, Ran-dependent mechanism. At the nucleoplasmic side of the NPC, GTP-Ran binds to importin beta and the three components separate, leading to release of the cargo. Importin alpha and beta are re-exported from the nucleus to the cytoplasm where GTP hydrolysis releases Ran from importin beta. The directionality of nuclear import is thought to be conferred by an asymmetric distribution of the GTP- and GDP-bound forms of Ran between the cytoplasm and nucleus. Mediates the nuclear import of histones H2A and H2B. Mediates the nuclear import of transcription factor GCN4.
Gene References into Functions
  1. Data suggest that, in nuclear exosomes, karyopherins are bound to the Rrp6 complex; Srp1, Kap95, and Sxm1 appear to be the most important karyopherins for nuclear import of Rrp6. (Rrp6 = exosome nuclease subunit RRP6; Srp1 = karyopherin alpha kap60; Kap95 = karyopherin beta Kap95; Sxm1 = karyopherin Kap108) PMID: 28539363
  2. The Kap95 is able to mediate efficient Mig2-Kap95 interaction in the absence of the importin-alpha. PMID: 23131016
  3. Hxk2 as a new cargo for the alpha/beta-importin pathway of S. cerevisiae. PMID: 22157003
  4. Kap95 is likely the primary karyopherin responsible for Replication protein A nuclear import in yeast. PMID: 21332387
  5. The authors identified karyopherin Kap95 as the transport factor responsible for the nuclear import of Slt2 and Rlm1. PMID: 19930464
  6. identification of a key interaction site where the RanGTP switch I loop binds to the carboxy-terminal arch of Kap95p PMID: 15864302
  7. There is a correlation between the binding affinity of an NLS cargo for the NLS receptor, importin alpha, and the import rate for this cargo. PMID: 16785238
  8. Kap95 serves as a key regulator of nuclear pore complex assembly into intact nuclear envelopes. PMID: 17182855
  9. Data show the crystal structure of the Kap95p-RanGDP complex shows that Kap95p induces the switch I and II regions of RanGDP to adopt a conformation that resembles that of the GTP-bound form. PMID: 18708071
  10. Pct1 interacts with Kap60 and Kap95 with this tripartite complex transiting the nuclear pore. PMID: 19141610
