Target Background

Function
ETS transcription factor with a prominent role during development of the eye and the nervous system. Required for glial-neuronal cell interactions at the ventral midline which are necessary for the proper elaboration of commissures in the embryonic CNS.; Required for normal EGFR-induced photoreceptor development probably as a downstream effector of Ras85D. In larval eye imaginal disks, activated by MAPK phosphorylation following EGFR activation, induces transcription of isoform P1 which in turn activates transcription of target genes essential for photoreceptor development.; Required for normal EGFR-induced photoreceptor development. Following transcriptional activation by isoform P2, acts as a constitutive activator of transcription, leading to induction of target genes essential for photoreceptor development. In larval brains, involved in the maintenance of type II neuroblast self-renewal and identity together with brat, btd and pros; prevents intermediate neuronal progenitor (INP) dedifferentiation by regulating the expression of erm probably via Notch signaling; suppresses Ase expression in type II neuroblasts and promotes the generation of intermediate neuronal progenitors.
Gene References into Functions
  1. results demonstrate that PntP1 prevents both the premature differentiation and the dedifferentiation of INPs by regulating the expression of distinct target genes at different stages of imINP development. PMID: 27510969
  2. Both PntP1 and PntP2 are required for oenocyte specification. Since PntP1 is a downstream effector of EGF signaling, these findings provide insight into how a Hox factor can both trigger and potentiate the EGF signal to promote an essential cell fate along the body plan. PMID: 28715417
  3. PntP2 transcription factor plays a role in air sac development by regulation of the EGF ligand Vein protein. PMID: 26632449
  4. Specific disruption of either pntP1 or pntP2 resulted in the same R8-only phenotype, demonstrating that both Pnt isoforms are essential for photoreceptor recruitment PMID: 23757412
  5. Demonstrate that the Pointed-P2 PNT domain contains a dynamic N-terminal helix H0 appended to a core conserved five-helix bundle diagnostic of the SAM domain fold. PMID: 22936607
  6. The MAPK/ERK pathway acts via the ETS-1 transcription factor Pointed. This mechanism permits physiological adjustment of insulin sensitivity and subsequent maintenance of circulating glucose at appropriate levels. PMID: 22242005
  7. Pointed directly regulates the transcription of string and thereby cell proliferation PMID: 12447387
  8. In different types of glial cells, REPO can act alone, or cooperate with either TTK69 or PNTP1 to regulate different target genes. PMID: 12702656
  9. Data show that pannier acts upstream of pointed P2 in a developmental pathway in which pannier promotes cardiac mesoderm formation, and pointed acts subsequently to distinguish between cardioblast and pericardial cell fates. PMID: 12756183
