Target Background

Function
DNA-dependent RNA polymerase (RNAP) catalyzes the transcription of DNA into RNA using the four ribonucleoside triphosphates as substrates.; Resistance to the antibiotics salinamide A, salinamide B, rifampicin, streptolydigin, CBR703, myxopyronin, and lipiarmycin can result from mutations in this protein.; Part of the processive rRNA transcription and antitermination complex (rrnTAC). The complex forms an RNA-chaperone ring around the RNA exit tunnel of RNAP. It supports rapid transcription and antitermination of rRNA operons, cotranscriptional rRNA folding, and annealing of distal rRNA regions to allow correct ribosome biogenesis.
Gene References into Functions
  1. The C-terminal segment of coliphage HK022 Nun interacts with the RNA polymerase beta and beta' subunits inside the RNA polymerase active site cleft as well as with nearly every element of the nucleic acid scaffold, essentially crosslinking the RNA polymerase and the nucleic acids to prevent translocation. PMID: 28318486
  2. The properties of generated RNA polymerase (RNAP) beta subunit variants Iindicate an RNA/protein interaction network that is crucial for transcription, thereby explaining the details of initiation-to-elongation transition on atomic level. PMID: 29036608
  3. The D654Y single amino acid substitution in RpoB rendered E. coli cells resistant to osmotic stress, probably due to improved cell growth and viability via enhanced sugar uptake under stressed conditions, and activated a potential "pre-defense" mechanism under non-stressed conditions. The findings of this work will be useful for bacterial host improvement to enhance its resistance to osmotic stress and facilitate bio-base PMID: 28193207
  4. Genotype-by-environment interactions of rpoB generated by antibiotic resistance mutations are more pronounced after adaptation to other types of environmental variation, making it difficult to predict long-term selection PMID: 23701170
  5. analysis of RNA polymerase rpoB mutations that alter transcription slippage during elongation in Escherichia coli PMID: 23223236
  6. data suggest that contact of the RNA polymerase beta subunit flap-tip helix hydrophobic patch to the sigma region 4 hydrophobic core is essential for stable interaction of the flap-tip helix with region 4 PMID: 15465046
  7. Amino acid substitutions define a highly conserved surface-exposed patch on the beta1 domain of RNA polymerase beta subunit that is required for the initial step of transcription-coupled repair. PMID: 15687384
  8. dG1551 and dC1692 in rpoB are critical for rifampicin drug resistance. PMID: 16276894
  9. Data report the structure of Escherichia coli GreB, and show that the carboxy-terminal coiled-coil of the RNA polymerase beta'-subunit is the main binding site for Gre factors. PMID: 17917675
  10. RNA polymerase is able to bind and react with many different RNAs and we suggest that RNAs are involved in transcriptional regulation more frequently than anticipated. PMID: 18388495
