Description

To create the US12 antibody, a rabbit is immunized with recombinant herpes simplex virus type 2 ICP47 protein (1-78aa), leading to the production of tailored IgG antibodies by the rabbit's B lymphocytes. The polyclonal US12 antibody is then subjected to rigorous purification using protein G techniques. This US12 antibody has been validated for its effective recognition of herpes simplex virus type 2 ICP47 protein in both ELISA and WB assays.

Herpes simplex virus type 2 (HSV-2) ICP47 protein shares only 42% amino acid sequence identity with HSV-1 ICP47. HSV-2 ICP47 protein plays a critical role in immune evasion by the virus. ICP47 specifically targets the antigen-dependent transporter (TAP) and disrupts its function, preventing the transport of viral antigens into the endoplasmic reticulum. This interference with antigen presentation inhibits the host immune response, allowing the virus to evade immune detection and clearance.

Function
Plays a role in the inhibition of host immune response. Binds specifically to transporters associated with antigen processing (TAP), thereby blocking peptide-binding and translocation by TAP as well as subsequent loading of peptides onto MHC class I molecules. Empty MHC I molecules are retained in the endoplasmic reticulum and ultimately directed to proteasomal degradation. In consequence, infected cells are masked for immune recognition by cytotoxic T-lymphocytes.
