Target Background

Function
Toxic component of a type II toxin-antitoxin (TA) system. A sequence-specific mRNA endoribonuclease that inhibits translation elongation and induces bacterial stasis. Cleavage occurs between the second and third residue of the Lys codon followed by a G or A (5'AAA(G/A)3'), is reading-frame dependent and occurs within the 5' end of most mRNAs. Ribosome-binding confers the sequence specificity and reading frame-dependence. When overexpressed in liquid media YafQ partially inhibits protein synthesis, with a reduction in growth rate and colony growth rate. This effect is counteracted by coexpression with cognate antitoxin DinJ. YafQ and DinJ together bind their own promoter, and repress its expression.; Cell death governed by the MazE-MazF and DinJ-YafQ TA systems seems to play a role in biofilm formation.
Gene References into Functions
  1. The findings demonstrate the plasticity of ribosome-dependent toxin active site residues by the E. coli YafQ toxin and further our understanding of which toxin residues are important for function. PMID: 26261214
  2. YafQ increases persistence by reducing indole, and toxin/antitoxin systems are related to cell signalling. PMID: 25041421
  3. The endoribonuclease activity of YafQ was inhibited in the complex with DinJ antitoxin in vitro. PMID: 22247505
  4. Acts as toxin in active toxin-antitoxin module with DinJ. Structure modelling suggests that it might act as a sequence-specific mRNA endoribonuclease. PMID: 17263853
  5. YafQ endoribonuclease expression resulted in translation, but not transcription or replication defects. PMID: 19210620
