Precision
Intra-assay Precision (Precision within an assay): CV%<8%
Three samples of known concentration were tested twenty times on one plate to assess.
Inter-assay Precision (Precision between assays): CV%<10%
Three samples of known concentration were tested in twenty assays to assess.

Linearity
To assess the linearity of the assay, samples were spiked with high concentrations of bovine Hpt in various matrices and diluted with the Sample Diluent to produce samples with values within the dynamic range of the assay.

Description

The bovine haptoglobin (HP) ELISA Kit is engineered for accurate measurement of bovine haptoglobin levels from samples including serum or plasma. It uses the cmpetitive-ELISA mechanism in combination with the enzyme-substrate chromogenic reaction to measure the bovine haptoglobin content in the sample. The color intensity is negatively correlated with haptoglobin content in the sample. This kit has been validated against standards of sensitivity, specificity, precision, linearity, recovery, and lot-to-lot consistency.

Haptoglobin is an acute-phase protein that is synthesized mainly in the liver and lungs and is crucial for the elimination of free hemoglobin and the neutralization of oxidative damage. It has powerful regulatory activity on lymphocytes. Additionally, haptoglobin exhibits immunoregulatory properties, participates in angiogenesis and inhibition of nitric oxide, stimulates tissue repair, etc.

 Q&A
Q:

a. Is the capture antibody a monoclonal or polyclonal?
b. What is the host of the capture antibody?
c. Is the detection antibody a monoclonal or polyclonal?
d. What is the host of the detection antibody?
e. What’s is that standard? A recombinant protein or something else? If a recombinant protein, what is the host?
f. If recombinant protein, is it full length or partial?
g. What is the species of the standard– human or something else?

A:
Thanks for your inquiry.
The antibody used in the kit is rabbit polyclonal antibody.
The standard is purified crude extract of native samples.

Target Background

Function
(From Uniprot)
As a result of hemolysis, hemoglobin is found to accumulate in the kidney and is secreted in the urine. Haptoglobin captures, and combines with free plasma hemoglobin to allow hepatic recycling of heme iron and to prevent kidney damage. Haptoglobin also acts as an antioxidant, has antibacterial activity and plays a role in modulating many aspects of the acute phase response. Hemoglobin/haptoglobin complexes are rapidly cleared by the macrophage CD163 scavenger receptor expressed on the surface of liver Kupfer cells through an endocytic lysosomal degradation pathway.
Gene References into Functions
  1. These results indicate that it is possible to monitor the course of pyometra in cows based on the evaluation of the concentration of cytokines and Hp in the serum and uterine washings. PMID: 25704413
  2. These data demonstrate that serum neutrophil haptoglobin-MMP 9 complexes appear sooner and decline more rapidly than other acute phase proteins. PMID: 25358728
  3. An increase in serum amyloid A or haptoglobin in dairy cows with abomasal displacementis not specific for inflammation or bacterial infection; increased levels of either may indicate the presence of hepatic lipidosis. PMID: 20391639
  4. Haptoglobin concentration may be a useful indicator for cows with postpartum reproductive disorders. PMID: 14960809
  5. Presence in the milk of the acute phase proteins (APP), haptoglobin (Hp) and serum amyloid A to see the relationships in cow composite and bulk tank milk samples. PMID: 17227596
  6. Bovine granulocytes, isolated from peripheral blood of healthy cattle, contain abundant amounts of haptoglobin within the granules. PMID: 17681384
  7. This study suggests that neutrophils and epithelial cells may play an essential role in elevating milk haptoglobin (Hp) in addition to previous suggestions that Hp may be derived from mammary tissues and circulation. PMID: 19094922
