This ELISA kit provides quantitative measurement of soluble gp130 (sgp130) in human samples, supporting research into cytokine signal transduction through the IL-6/JAK-STAT pathway. Because sgp130 functions as a natural inhibitor of IL-6 trans-signaling — a mechanism implicated in chronic inflammation and autoimmunity — precise quantification across both circulating and secreted fractions is essential for dissecting this regulatory axis.
Detection Range: The 0.24–15 ng/ml range aligns with reported circulating sgp130 concentrations in human serum and plasma, which typically fall between 100–400 ng/ml, meaning most serum samples will require dilution. Conditioned media from stimulated cells, where sgp130 shedding may be lower, can often be measured directly within this range.
Sensitivity: At 0.06 ng/ml, the kit detects sgp130 in low-abundance matrices such as saliva, urine, and conditioned media from unstimulated or primary cell cultures where shed gp130 levels are minimal.
Sample Compatibility: Validation across serum, plasma, cell culture supernatants, saliva, and urine enables researchers to correlate systemic sgp130 levels with local shedding at mucosal or renal sites, bridging in vivo and in vitro trans-signaling studies within a single assay platform.
Assay Efficiency: A 50–100 µl sample requirement and 1–5 hour protocol accommodate limited primary cell supernatants and high-throughput screening of patient cohorts.
