CUSABIO's IGF2 human ELISA kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of human IGF2 in multiple samples, including serum, plasma, cell culture supernates, urine, and tissue homogenates. This assay uses an antibody specific to human IGF2 coated on a 96-well plate. IGF2 present in a sample is bound to the wells by the immobilized antibody after adding the sample to the wells. Biotinylated IGF2 antibody is added to the wells, forming an antibody-analyte-antibody complex. HRP-conjugated avidin is pipetted to the wells and binds to the complex. A TMB substrate solution is added to the wells and color develops in proportion to the amount of IGF2 bound. The addition of Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm using a microplate reader.
IGF2 is a protein hormone that regulates cell proliferation, growth, migration, differentiation, and survival. IGF2 binds to IGF2R and is targeted for degradation, while engagement of IGF2 to IGF1R stimulates signaling pathways involved in metabolism, proliferation, differentiation, and anti-apoptosis. It also plays an essential role in bone development, where it promotes the proliferation and differentiation of bone cells. IGF2 has been proposed to participate in the development of various seemingly unrelated cancers that play an important role in geriatric medicine, such as breast cancer, colon cancer, and lung cancer. IGF2 has also been involved in cardiovascular diseases.
