The Rat C-telopeptide-of-type I (CTX-I) ELISA kit is a ready-to-use microwell, strip plate ELISA kit for the quatitative measurement of rat CTX-I in biological samples, including serum, plasma, and tissue homogenates. It empolys the competitive ELISA technique in the detection of target analytes in the samples. This ELISA kit has been quality controlled in multiple characterisitics, including sensitivity, specificity, precision, linearity, and recovery. Standards or samples are added to the appropriate microtiter plate wells pre-coated with goat-anti-rabbit CTX-I antibody. HRP-conjugated CTX-I and anti-rat CTX-I antibody are pipped into the wells, where CTX-I in the sample or standards competes with the HRP labeled CTX-I for limited binding sites on the anti-rat CTX-I antibody. these Ag-Ab complexes are captured by the antibody immobilized on the plate. The solution color turns blue after the addition of substrate A and B. The color reaction is immediately terminated after the addition of stop solution. And the color changes from blue to yellow. The color develops oppositely to the amount of CTX-I in the sample, and the intensity of the color is measured by Microplate reader at 450 nm.
CTX-I is a sensitive and variable marker of bone resorption. Shereen M Olama etc, showed that serum CTX-I level was significantly elevated in patients with fibromyalgia syndrome (FMS) and that increased experssion of CTX-I was associated with the disease severity. Increased CTX-I may cause the early development of osteoporosis.

Recovery |

- A 96-well Assay plate -- The 96-well plate has been pre-coated with goat-anti-human CTX-I antibody.
- Standard 6 x 0.5 ml -- Dilute the standard at dilution series, read the OD values, and then draw a standard curve.
- Anti-rat CTX-I Antibody 1 x 6 ml -- Act as the detection antibody.
- HRP-conjugated CTX-I 1 x 6 ml -- Bind to the CTX-I antibody, and HRP catalyzes the substrate A and B to elicit a chromogenic reaction.
- Wash Buffer (20 x concentrate) 1 x 15 ml -- Wash away unbound or free substances.
- Substrate A 1 x 7 ml -- Mix with substrate B, and the TMB is catalyzed by HRP to develop color.
- Substrate B 1 x 7 ml -- Mix with substrate A, and the TMB is catalyzed by HRP to develop color.
- Stop Solution 1 x 7 ml -- Stop the color reaction. The solution color immediately turns from blue to yellow.
