Experimental Principle

Antibodies are bound to the corresponding proteins in the lysate, then incubated with ProteinA/G coupled Sepharose or MagneticBeads, and the ProteinA/G bead-antibody-destination protein complex is obtained by centrifugation or with the help of a magnetic rack. Under the action of high temperature and reducing agent, the antigen and antibody are dissociated, and the supernatant is collected, which includes the antibody, destination protein and a small amount of heteroprotein. The supernatant includes antibody, target protein and a small amount of heterogeneous protein, and then the protein is identified by Western Blot or mass spectrometry (MS).
