Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Description

IL4R mediates the pleiotropic effects of interleukin-4 in immune regulation and allergic inflammation, making its extracellular domain a critical target for therapeutic antibody development and mechanistic studies of cytokine signaling. This mammalian-expressed fragment spanning residues 26–232 captures the ligand-binding domain with native glycosylation patterns that support physiologically relevant interactions, as demonstrated by functional ELISA showing an EC50 of 12.68–14.23 ng/mL when binding immobilized IL-4. The quantified binding affinity provides a validated basis for competitive inhibition assays, blocking antibody screening, and therapeutic antibody epitope mapping studies where accurate receptor-ligand kinetics are essential. Receptor for both interleukin 4 and interleukin 13. Couples to the JAK1/2/3-STAT6 pathway. The IL4 response is involved in promoting Th2 differentiation. The IL4/IL13 responses are involved in regulating IgE production and, chemokine and mucus production at sites of allergic inflammation. In certain cell types, can signal through activation of insulin receptor substrates, IRS1/IRS2.; Soluble IL4R (sIL4R) inhibits IL4-mediated cell proliferation and IL5 up-regulation by T-cells.
