IL-4R, consisting of 802 amino acid residues and weighing 140kDa, belongs to the erythropoietin receptor superfamily. Its structure is characterized by specific features, including two fibronectin type III domains (fnIII), each composed of six β-sheets. These domains contribute to the formation of heterodimers when IL-4 binds to the IL-4Rα subunit. IL-4R plays a pivotal role in numerous cellular processes. It is involved in the survival and development of immune cells, regulating the development and maturation of B cells, as well as the proliferation and differentiation of type II helper T cells. Additionally, the expression of IL-4R is closely associated with the survival, proliferation, migration, and invasion of various tumor cells. In the tumor microenvironment, the expression of IL-4R is upregulated and promotes tumor metastasis through multiple mechanisms. The biological functions of IL-4R are primarily mediated through the activation of the non-receptor tyrosine kinase JAK. When IL-4 binds to IL-4R, it activates JAK, triggering a series of signaling cascades. These reactions include the phosphorylation and nuclear translocation of STAT6, as well as the activation of other downstream signaling molecules. These signaling pathways ultimately affect cellular processes such as proliferation, differentiation, survival, and migration.
