Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Involved in RNA-mediated post-transcriptional gene silencing (PTGS). Main component of the RNA-induced silencing complex (RISC) that binds to a short guide RNA such as a microRNA (miRNA) or small interfering RNA (siRNA). RISC uses the mature miRNA or siRNA as a guide for slicer-directed cleavage of homologous mRNAs to repress gene expression. Required for reliable formation of primary and axillary shoot apical meristems. Specifies leaf adaxial identity by repressing the miR165 and miR166 microRNAs in the embryonic shoot apex, in the shoot apical meristem (SAM) and leaf. Represses the microRNA miR398 which targets CCS1 chaperone mRNAs for translational inhibition. Acts as a negative regulator of AGO1 protein level. Like AGO1, is required for stem cell function and organ polarity. Unlike AGO1, is not subjected to small RNA-mediated repression itself. Essential for multiple processes in development. Coregulates, with GATA18/HAN, the shoot apical meristem (SAM) organization.
Gene References into Functions
  1. the shoot regeneration inhibition determined by AGO10 functions via the repression of miR165/166. PMID: 29016889
  2. The regulatory mechanism of AGO10 by which shoot apical meristem size is controlled under stress conditions.[LBD12-1] PMID: 27895202
  3. data suggest that boundary-expressing HAN communicates with the meristem through the PNH, regulates floral organ development via JAG and BOP2, and maintains boundary morphology through CKX3 during flower development in Arabidopsis PMID: 26390296
  4. Our results indicate that maintenance of shoot meristem stem cells by AGO10 involves negative regulation of auxin signaling and, via REV-mediated downregulation of ARF2 expression, auxin response. PMID: 26358077
  5. AGO1, AGO2 and AGO10 promoted anti-TuMV defense in a modular way in various organs, with AGO2 providing a prominent antiviral role in leaves. AGO5, AGO7 and AGO10 had minor effects in leaves. PMID: 25806948
  6. AGO10 quenches miR165/166 that move into AGO10-expressing niches and promotes shoot apical meristem maintenance. PMID: 25801022
  7. AGO10 specifies leaf adaxial identity by repressing miR165/166. PMID: 19054365
  8. ubiquitously expressed ARGONAUTE1 (AGO1) gene and its homolog AGO10, which exhibits highly restricted spatial expression patterns, are required to maintain the correct temporal program of floral stem cells PMID: 21483759
  9. Using an unbiased biochemical approach, we show that AGO10 specifically binds to miR166/165. This association is determined by distinct structural features of the miR166/165 duplex. PMID: 21496644
  10. proposed that ZWILLE buffers meristem stability non-cell-autonomously by ensuring the critical number of apical cells required for proper meristem function PMID: 14564520
  11. Analysis of ago1 mutants with partially compromised AGO1 activity revealed that loss of ZLL function re-establishes both siRNA and miRNA pathways for a subset of AGO1 target genes. PMID: 19763164
