Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Involved in cadmium/zinc transport.
Gene References into Functions
  1. To test whether developing Arabidopsis seeds depend on AtHMA2 and AtHMA4 for proper zinc unloading from the seed coat, the authors analysed the amount of zinc present in the maternal and filial parts of the seed, respectively, in wild-type and mutant seeds lacking functional AtHMA2, AtHMA4, or both. PMID: 27243644
  2. overexpression modulates the ratio of Zn to Cd supply as a determinant of metal-homeostasis gene expression in tobacco PMID: 27811086
  3. These studies evaluated the the function and structure of the N-terminal cytoplasmic metal-binding domain of HMA4. PMID: 26797794
  4. Due to ectopic expression of 35S::AtHMA4, the physical apoplastic barrier within the external cell layer developed, which is likely to be responsible for the reduction of Cd uptake/accumulation. PMID: 24420575
  5. Ectopically expressing AtHMA4 in tobacco results in overloading the root and leaf apoplast with zinc. PMID: 23170996
  6. the C-terminal domain of AtHMA4 serves a dual role as Zn(2+) and Cd(2+) chelator (sensor) and as a regulator of the efficiency of Zn(2+) and Cd(2+) export. PMID: 20650903
  7. The binding affinities of HMA2 and HMA4 for zinc are higher than that of HMA7 by a factor of 20-30, but the relative affinities for copper are inverted by a factor of 30-50. PMID: 19883117
  8. AtHMA4 overexpressing lines displayed an increase in the zinc and cadmium content of shoots. [AtHMA4] PMID: 15498553
  9. Mutagenesis of the cysteine and glutamate residues from the N-ter degenerated heavy metal binding domain impaired the function of AtHMA4 [HMA4] PMID: 15733866
  10. The near-complete abolition of root-to-shoot Cd translocation resulting from the loss of function of HMA2 and HMA4 demonstrates they are the major mechanism for Cd translocation in Arabidopsis thaliana. PMID: 19076718
