Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Casein kinases are operationally defined by their preferential utilization of acidic proteins such as caseins as substrates. Can phosphorylate a large number of proteins. Negative regulator of wg signaling. Phosphorylates arm directly or indirectly and stimulates its degradation which prevents inappropriate wg signaling. Phosphorylates smo which promotes its accumulation at the cell surface and its signaling activity in response to hh. Together with dco, regulates proteolytic processing of ci by phosphorylating it which promotes its binding to slmb, the F-box recognition component of the SCF(slmb) E3 ubiquitin-protein ligase required for ci processing. Inhibits condensin II interphase activity by promoting degradation of the Cap-H2 regulatory subunit and limiting the levels of chromatin-bound Cap-H2 which regulates interphase chromosome organization.
Gene References into Functions
  1. glucose regulatory role PMID: 25994086
  2. we identify Casein Kinase I alpha (CK1alpha) as an additional negative-regulator of Cap-H2. CK1a-depletion stabilizes Cap-H2 protein and results in an accumulation of Cap-H2 on chromosomes. PMID: 25723539
  3. Expected genes in the Scar-Arp2/3 actin nucleation pathway were identified as well as casein kinase I, which had a similar morphological RNAi signature. PMID: 21041442
  4. CKI(Casein kinase I) functions as a negative regulator of Arm protein, by phosphorylating it on Ser and Thr residues in the N-terminus and targeting it for degradation. PMID: 11927557
  5. 2 proteins reported to function in Wingless(Wg) signaling identified as Hh pathway components: Dally-like protein required for Hh signal reception, and casein kinase 1alpha, a candidate tumor suppressor that regulates activities of both Hh and Wg pathways PMID: 12663920
  6. expression of doubletime in Drosophila S2 cells produced a CKI-7-sensitive kinase activity which was reduced by both the dbt(S) and dbt(L) mutations of circadian rhythm PMID: 14701759
  7. doubletime and CKII kinases collaborate to potentiate Drosophila PER transcriptional repressor activity PMID: 14759367
  8. Several CKI isoforms including CKIalpha and Gish/CKIgamma can phosphorylate the Wg coreceptor Arrow (Arr), which may account, at least in part, for their positive roles in the Wg pathway. PMID: 16987508
