Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Receptor for ecdysone. May be an important modulator of insect metamorphosis. Plays an important part in embryonic and post-embryonic development. Binds to ecdysone response elements (ECRES) such as in the promoter region of s15 chorion gene.
Gene References into Functions
  1. Context-dependent functional properties of EcR and USP receptors in Drosophila cell survival. PMID: 26250909
  2. nutrition controls ecdysone biosynthesis partially via FoxO-Usp prior to critical weight, ensuring that growth only stops once larvae have achieved a target nutritional status. PMID: 25421296
  3. The results suggest that normal larval to pupal maturation in this mecopteran model insect requires the involvement of a distinct endocrine axis of USP binding to its own endogenous terpenoid ligand. PMID: 23211750
  4. PKC mediates USP phosphorylation at Ser35 in Kc and S2 cells, which regulates 20-hydroxyecdysone signaling. PMID: 23136906
  5. sumoylation of Usp protein can be an important factor in modulating its activity by changing molecular interactions PMID: 22676916
  6. Ecdysone and NO mediate gene regulation by competing EcR/Usp and E75A nuclear receptors during Drosophila development PMID: 21981918
  7. Ecdysone receptor and USP were expressed in numerous adult tissues, but receptor activity varied depending on tissue type and adult age. Receptor activity is reduced when a constitutively inactive ecdysone receptor is present. PMID: 21507325
  8. Data show that ultraspiracle- and ecdysteroid receptor-mediated transcriptional activity not only depends on an interaction between the ecdysteroid receptor and a given hormone response element, but is also modified by surrounding DNA sequences. PMID: 21316451
  9. stability of the ecdysone receptor (EcR) is regulated in a cooperative manner by the AB-domains of EcR and Ultraspiracle PMID: 19711356
  10. The heterodimerization partner Usp increased DNA binding of ecdysone receptor considerably PMID: 19750549
  11. USP both represses and activates a gene affecting furrow movement, the ecdysone-responsive Z1 isoform of Broad-Complex, and we report additional usp mutant phenotypes PMID: 11804781
  12. ecdysteroids are capable of inducing heterodimer formation between EcR and USP PMID: 12084064
  13. USP activates transcription and binds ligand with a functional outcome of enhanced promoter activity PMID: 12473098
  14. truncation of its N-terminal A/B region changes molecular characteristics of ultraspiracle, considerably weakening its oligomerization potential under the same experimental conditions PMID: 12674500
  15. findings show that the EcR-USP ecdysone receptor is first activated in the extraembryonic amnioserosa, implicating this tissue as a source of active ecdysteroids in the early embryo PMID: 12958367
  16. Analysis of the role of ligand-binding domains in the heterodimerization of ecdysone receptor and ultraspiracle protein. PMID: 14594447
  17. results indicate that EcR and Usp can enter the nucleus independently, but cotransport of both receptors mediated by dimerization via the ligand binding domains is possible even in the absence of hormone PMID: 17570142
  18. data confirm that EcR and Usp can enter the nucleus independently and that intracellular localization is regulated individually for each receptor PMID: 17570491
  19. A bimolecular fluorescence complementation (BiFC) method was used to directly analyze the formation of the EcR/Usp complex. PMID: 18771703
