Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.

Target Background

Function
Involved in the regulation of nitrogen metabolism. Regulates the activity of its targets by protein-protein interaction in response to the nitrogen status of the cell. Involved in the regulation of the ammonium transporter AmtB so as to optimize ammonium uptake under all growth conditions. In nitrogen-limited conditions, GlnK does not interact with AmtB, which remains active and imports ammonium. When extracellular ammonium increases, GlnK associates tightly with AmtB in the inner membrane, thereby inhibiting the transporter activity. Also involved in the regulation of the glutamine synthetase adenylyltransferase/adenylyl-removing (AT/AR) enzyme GlnE and the glutamine synthetase GlnA. In nitrogen-limited conditions, formation of uridylylated GlnB(PII)/GlnK heterotrimers may fine-regulate the activation of GlnA: uridylylated heterotrimers stimulate the deadenylation and the activation of GlnA whereas uridylylated GlnK homotrimers do not stimulate, or hardly stimulate, the deadenylation of GlnA. In addition, regulates the expression of Ntr genes during nitrogen starvation, probably via the control of the levels of phosphorylated NRI during different stages of the response to nitrogen limitation.

  1. Results show that GlnK is tightly regulated under nitrogen-rich conditions, yet it is expressed during ammonium run-out and starvation. This suggests a role for GlnK as a buffer of nitrogen shock after starvation, and provides a further functional link between nitrogen and carbon metabolisms. PMID: 28538158
  2. Data show that the P(II) protein, GlnK, has an ATPase activity that is inhibited by 2-oxoglutarate. PMID: 23818625
  3. Studies suggest a hypothesise that the regulatory protein GlnK is required to fine-tune the active transport of ammonium in order to limit futile cycling whilst enabling an intracellular ammonium level sufficient for the cell's nitrogen requirements. PMID: 21134373
  4. Co-purification of GlnK with AmtB from ammonium-shocked E. coli cells definitvely demonstrates that these two proteins form a functional complex with a stoichiometry of 1:1 and dependent on the uridylylation status of GlnK. PMID: 16864585
