To prepare this Recombinant HEV-1 ORF2 protein, the recombinant DNA was required, which was generated by fusing the ORF2 gene with N-terminal 6xHis-SUMO tag sequence. Once the recombinant DNA was amplified and purified, a protein expression system, E.coli, was needed for this ORF2 protein production. After purification, a premium ORF2 recombinant proteinwas obtained. According to SDS-PAGE, its purity turns out to be 90%+.
ORF2 is a protein encoding gene that provides an instruction in making a protein named secreted protein ORF2 or protein ORF2. In Hepatitis E virus (HEV), the protein encoded by ORF2 gene has two isoforms, secreted protein ORF2 and capsid protein ORF2. The sequence difference between isoform 2 and the isoform 1 is lack of the 1-15 amino acid sequence. The difference between the sequence directly determines their different functions. Isoform 1 can inhibit host antibody-mediated neutralization without blocking viral cell entry. Isoform 2 is the major virion component to form the viral capsid.
