The preparation of this recombinant Human TPH2 protein was to use gene recombination DNA technology to obtain a recombinant vector connected with a TPH2 fragment (1-490aa) that could be translated into the TPH2 protein and then transferred it into E.coli cells to express the recombinant TPH2 protein molecule. In order to get the target protein with high purity, N-terminal 6xHis tag was used in the production. The purity is 85% determined by SDS-PAGE.
Tryptophan hydroxylase 2 (TPH2) is the rate-limiting enzyme in brain serotonin synthesis. The TPH2 gene has frequently been investigated in relation to psychiatric morbidity. Thus, TPH2 plays a pivotal role in the regulation of food intake and body weight. Mutations in this TPH2 may be associated with psychiatric diseases such as bipolar affective disorder and major depression. The central 5-hydroxytryptamine system has long been explored for its potential as a pharmacological target for obesity. Altered function of tryptophan hydroxylase-2 (TPH2), the enzyme critical for synthesis of serotonin in the brain, plays a role in anxiety-, aggression- and depression-related personality traits and in the pathogenesis of disorders featuring deficits in cognitive control and emotion regulation.
