Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Serine/threonine kinase which responds to activation by environmental stress and pro-inflammatory cytokines by phosphorylating downstream targets. Phosphorylates skn-1 which probably regulates skn-1 nuclear translocation in response to oxidative stress. Probably by activating skn-1, involved in the up-regulation of gcs-1 and glutathione-S-transferase gst-4 expression upon bacteria infection. Up-regulates expression of gcs-1 in intestinal cells upon arsenite treatment. Functions downstream of the MAPKK sek-1 and the MAPKKK nsy-1 as the MAP kinase which regulates pathogen resistance and responses to oxidative stress. Required for expression of antimicrobial peptide nlp-29 in response to fungal infection or physical injury. Involved in resistance to the nematotoxic C.cinerea galectin Cgl2. May play a redundant role with other MAP kinases in susceptibility to anoxia, downstream of tir-1/nsy-1. Phosphorylates transcription factor rnt-1 during oxidative stress which results in rnt-1 stabilization in the intestine. Phosphorylates transcription factor atf-7 during pathogen infection resulting in modulation of target genes. Probably downstream of nsy-1 and sek-1, involved in germline apoptosis induced by heavy metals, such as Cu(2+).
Gene References into Functions
  1. The authors used RNAi to knock down p38 MAPK (pmk-1) expression. C. elegans MAPK mutants are hypersusceptible to mycobacterial infection and pathogenic mycobacteria inhibit MAPK-mediated protection through the MAPK phosphatase gene. PMID: 26919641
  2. PMK-1 plays an important protective role in silver nanoparticle-induced oxidative DNA damage-repair. PMID: 24347047
  3. the PMK-1 pathway is involved in the heat stress responses of C. elegans. PMID: 23117578
  4. define the phenomenon of innate immunosenescence in C. elegans in terms of the age-dependent dynamics of the PMK-1 innate immune signaling pathway, and they suggest that a cycle of intestinal tissue aging, immunosenescence, and bacterial proliferation PMID: 21625567
  5. These results establish a PMK-1-dependant PCD pathway as a C. elegans innate immune response to Salmonella PMID: 12526744
  6. The contribution of the PMK-1 pathway to the enhanced lifespan of daf-2 mutants suggests that innate immunity is an important determinant of longevity. PMID: 17096597
