The recombinant mouse 1-phosphatidylinositol 4,5-bisphosphate phosphodiesterase delta-1 (Plcd1) is produced using a baculovirus expression system. The transfer plasmid contains the full-length mouse Plcd1 protein, spanning amino acids 1 to 756, along with a C-terminal 6xHis-tag for purification. Insect cells are co-transfected with a mixture of the transfer plasmid and baculovirus DNA construct, facilitating the generation of the recombinant baculovirus particles. After that, infecting insect cells with the recombinant baculovirus carrying the Plcd1 expression construct. The infected cells then produce and accumulate the recombinant Plcd1 protein. The purified recombinant Plcd1 protein exhibits a purity of up to 85%, as determined by SDS-PAGE analysis. On the gel, the Plcd1 protein migrates as a band with an approximate molecular weight of 92 kDa.
Plcd1 is involved in various cellular processes, including intracellular calcium signaling, regulation of ion channels, and modulation of the protein kinase C (PKC) signaling pathway. By regulating the levels of IP3 and DAG, Plcd1 contributes to the dynamic control of cellular responses, such as gene expression, cytoskeletal rearrangement, and cell proliferation.
