Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol. If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
High-affinity, saturable copper transporter involved in dietary copper uptake.
Gene References into Functions
  1. Gene duplication and neo-functionalization in the evolutionary and functional divergence of the metazoan copper transporters Ctr1 and Ctr2 PMID: 28507097
  2. studies identify a new processing event and the key protease that cleaves the Ctr1 metal-binding ectodomain, which functions to regulate cellular Cu(+) and cisplatin acquisition PMID: 27143361
  3. CTR1, ATP7A, and lysyl oxidase were upregulated in the lung tissues and pulmonary arteries of mice with hypoxia-induced pulmonary hypertension and pulmonary arterial smooth muscle cells. PMID: 24614111
  4. conclude that Y103 is required for the internalization of hCTR1 in response to Cu, that this occurs by a mechanism other than phosphorylation and that mutation of Y103 modulates the interaction with IRS-4 PMID: 24967972
  5. A key regulatory mechanism for mammalian copper transport is through Ctr2-dependent accumulation of a Ctr1 variant lacking the copper- and cisplatin-binding ecto-domain. PMID: 24167251
  6. conclude that Cu acquired from CTR1 is required for signaling in pathways regulated by RTKs that play major roles in development and cance PMID: 22842628
  7. There was little difference in rates/kinetics of uptake of copper in the Ctr1+/+ and -/- cells. Endocytosis was not involved. PMID: 22354499
  8. basal expression of Ctr1 is regulated by HIF2alpha; however, the induction by hypoxia is a HIF2alpha-independent event PMID: 22684009
  9. Structure-functional organization of eukaryotic high-affinity copper importer CTR1 determines its ability to transport copper, silver and cisplatin PMID: 22670529
  10. Data show apical localization of Ctr1 in intestinal epithelia and suggest that increased Ctr1 apical localization in response to dietary copper limitation may represent an adaptive response to modulate Ctr1 availability at the site of copper absorption. PMID: 20699218
  11. Ctr1 is an important transport protein in the accumulation of silver in mammalian cells. PMID: 20569931
  12. Mouse cell lines lacking one or both mouse Ctr1 (mCtr1) alleles exhibit increased cisplatin resistance and decreased cisplatin accumulation in parallel with mCtr1 gene dosage PMID: 12370430
  13. These studies identify Ctr1 as the major factor driving intestinal Cu absorption in mammals. PMID: 16950140
  14. These studies support a model in which vertebrate Ctr1 functions as a key regulator of the differentiation capacity of both stem and progenitor cell populations. PMID: 17620605
  15. Deletion of hepatic Ctr1 reveals its function in copper acquisition and compensatory mechanisms for copper homeostasis. PMID: 19095764
  16. Ctr1 is mainly localized on the basolateral side of both proximal and distal tubular cells in mouse kidneys. PMID: 19144690
