Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Required for the establishment of the specialized cells of the pituitary gland and the nervous system. Involved in the development of interneurons and motor neurons in cooperation with LDB1 and ISL1. Acts as a transcriptional activator. Binds to and activates the promoter of the alpha-glycoprotein gene, and synergistically enhances transcription from the prolactin promoter in cooperation with Pou1f1/Pit-1.
Gene References into Functions
  1. conditional deletion of Shh in the anterior hypothalamus results in a fully penetrant phenotype characterised by a complete arrest of (Rathke's pouch) RP development, with lack of Lhx3/Lhx4 expression in RP epithelium. PMID: 28807898
  2. We discovered that expression of Cdkn1a is expanded dorsally in the pituitary primordium of both Lhx3 and Lhx4 mutants. PMID: 25668206
  3. Lhx3-LIM domains play critical roles in transactivation of the Isl1-Lhx3 complex by not only directing the assembly of the Isl1-Lhx3 complex but also recruiting coactivators to the complex. PMID: 26260513
  4. This treatment leads to activation of Lhx3 gene expression. PMID: 24183897
  5. LHX3 interacts with inhibitor of histone acetyltransferase complex subunits LANP and TAF-1beta to modulate pituitary gene regulation. PMID: 23861948
  6. The results of this study suggested that Lhx3/Chx10 medullary reticular formation neurons are involved in locomotion. PMID: 24027269
  7. Isl1-Lhx3, a LIM-complex mimicking fusion, induces a signature of motor neuron transcriptome & suppresses interneuron differentiation programs. An equimolar ratio of Isl1 & Lhx3 & the LIM domain of Lhx3 are crucial for this. PMID: 22343290
  8. LHX3 gene encodes a LIM-homeodomain transcription factor, which has essential roles in pituitary and nervous system development in mammals. PMID: 21149718
  9. LIM factor Lhx3 contributes to the specification of motor neuron and interneuron identity through cell-type-specific protein-protein interactions PMID: 12150931
  10. study demonstrates a functional role for LHX3 in the regulation of the GnRH receptor PMID: 15705775
  11. underscore the view that Lhx3 functions downstream of the Pitx factors in the same transcriptional cascade that controls growth and early cell differentiation of the developing pituitary gland PMID: 16859901
  12. These overexpression studies provide insights into the properties of LHX3 during pituitary development and highlight the importance of this factor in reproductive physiology. PMID: 17311285
  13. analysis of expression of LHX3 and SOX2 during mouse inner ear development PMID: 17604700
  14. the abnormal proteins displayed diminished capacities to activate the promoters of genes expressed in the pituitary gland. On nervous system promoters, several mutant proteins retained some activity PMID: 17616267
  15. Lhx3 contributes to pituitary development by maintaining normal dorsal-ventral patterning, cell survival, and normal expression of corticotrope-specific transcription factors, which are necessary for repressing ectopic gonadotrope differentiation. PMID: 18037398
  16. Results show that Lhx3 exhibits specific spatial and temporal expression in the mouse placenta, but that absence of both LHX3 and 4 genes did not interfere with placental transport nor consistently with expression of target genes such as Gnrhr. PMID: 18425848
  17. The second wave of expression at E12.5 includes Lhx3, 5, 9, Isl2, and Lmx1b in the differentiating sensory epithelia with cellular specificities. PMID: 18942141
