Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Transcription factor that binds a canonical ESRRB recognition (ERRE) sequence 5'TCAAGGTCA-3' localized on promoter and enhancer of targets genes regulating their expression or their transcriptional activity. Plays a role, in a LIF-independent manner, in maintainance of self-renewal and pluripotency of embryonic and trophoblast stem cells through different signaling pathways including FGF signaling pathway and Wnt signaling pathways. Upon FGF signaling pathway activation, interacts with KDM1A by directly binding to enhancer site of ELF5 and EOMES and activating their transcription leading to self-renewal of trophoblast stem cells. Also regulates expression of multiple rod-specific genes and is required for survival of this cell type. Plays a role as transcription factor activator of GATA6, NR0B1, POU5F1 and PERM1. Plays a role as transcription factor repressor of NFE2L2 transcriptional activity and ESR1 transcriptional activity. During mitosis remains bound to a subset of interphase target genes, including pluripotency regulators, through the canonical ESRRB recognition (ERRE) sequence, leading to their transcriptional activation in early G1 phase. Can coassemble on structured DNA elements with other transcription factors like SOX2, POU5F1, KDM1A and NCOA3 to trigger ESRRB-dependent gene activation. This mechanism, in the case of SOX2 corecruitment prevents the embryonic stem cells (ESCs) to epiblast stem cells (EpiSC) transition through positive regulation of NR0B1 that inhibits the EpiSC transcriptional program. Also plays a role inner ear development by controlling expression of ion channels and transporters and in early placentation.
Gene References into Functions
  1. The resulting cells express the components of the core transcriptional network of mESCs, including ESRRB, at high levels, which suggests the existence of naive-state hiPSCs that are similar to mESCs. We also demonstrate that these cells differentiate more readily into neural cells than do conventional hiPSCs. PMID: 29129686
  2. Taken together these observations suggest that ESRRbeta is a critical component in the onset of Ddilated cardiomyopathy by affecting contractility and calcium balance PMID: 28130335
  3. Findings indicate that the Gata6 promoter is activated by Esrrb in association with Ncoa3, and Dax1 inhibited activities of Esrrb and Ncoa3, resulting maintenance of the undifferentiated status of embryonic stem (ES) cells. PMID: 27601327
  4. mitotic Esrrb binding is highly dynamic, driven by specific recognition of its DNA-binding motif and is associated with early transcriptional activation of target genes after completion of mitosis PMID: 27723719
  5. JARID2 physically interacts with ESRRB, SALL4A, and PRDM14 PMID: 26523946
  6. interacts in trophoblast stem cells with the histone demethylase Lsd1 and with the RNA Polymerase II-associated Integrator complex PMID: 26206133
  7. The forced expression of Essrb (and Nanog) resulted in the dispensability of nucleostemin expression in embryonic stem cells. PMID: 25522312
  8. Daily regulation of Esrrb in photoreceptor cells may contribute to their adaptation to 24-h changes in metabolic demands. PMID: 25737630
  9. Loss of Errbeta affects body composition, neuropeptide levels, stress hormones, and centrally-modulated startle responses of mice. PMID: 24053666
  10. Esrrb selective deletion in the hindbrain alters neuropeptide Y expression, whole-body energy level and ingestive behavior. PMID: 23360481
  11. Dax1 functions as a negative regulator of Esrrb and Oct3/4, and these molecules form a regulatory loop for controlling the pluripotency and self-renewal capacity of embryonic stem cells. PMID: 23508100
  12. a direct Nanog target gene that can substitute for Nanog function in pluripotent cell PMID: 23040477

  14. Esrrb forms a heterodimer complex with Sox2 in ES cells to regulate transcriptional differences between mouse embryonic and epiblast stem cells. PMID: 23169531
  15. we present evidence that chondrolectin (Chodl) is expressed by fast motor neurons and that estrogen-related receptor beta (ERRbeta) is a candidate marker for slow motor neurons in the spinal cord PMID: 20437528
  16. ERRbeta is a critical regulator of rod photoreceptor function and survival PMID: 20534447
  17. Data demonstrate that estrogen-related receptor (ERR) beta is transiently induced, and ERRgamma is dramatically induced (and accumulates) in a differentiation-dependent manner in skeletal muscle cells. PMID: 19631715
  18. estrogen receptor beta knockout females exhibit lordosis behaviour equal to that of wild-type females PMID: 12969243
  19. ERbeta was predominantly localized in the magnocellular divisions of PVN. In the pregnant female brain. andcirculating sex steroids may differentially regulate the expression of ERbeta in the PVN of mice. PMID: 14715153
