Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Involved in the uptake of GABA and glycine into the synaptic vesicles.
Gene References into Functions
  1. Study investigated VIAAT expression during development in: 4 different wildtype-strains, 1 transgenic mouse line with congenital deafness and 1 mouse line with cochlear ablation. Results identify an activity-regulated gradient of VIAAT in the superior olivary complex of NRMI mice. Its absence in other mouse strains adds a novel layer of strain-specific features in the auditory system. PMID: 29073893
  2. GABAergic SVs that lacked the vesicular GABA transporter (VGAT) did not show the pH overshoot and acidified further to approximately 6.0. Comparison of luminal pH dynamics in the presence or absence of VGAT showed that VGAT operates as a GABA/H(+) exchanger, which is continuously required to offset GABA leakage PMID: 27601664
  3. The observed changes in gene and protein expression of vesicular GABA transporter, regional architecture, and morphology of mouse auditory forebrain may relate to-and to some extent enable-the emergence of mature sound-evoked activity patterns. PMID: 26159773
  4. the AD-like tau accumulation induces anxiety through disrupting miR92a-vGAT-GABA signaling, which reveals molecular mechanisms underlying the anxiety behavior in AD patients and potentially leads to the development of new therapeutics for tauopathies. PMID: 28129110
  5. This study demonstrated that Alterations in hypoglossal motor neurons due to VGAT deficiency in mice. PMID: 27956032
  6. The results suggest that vesicular release of GABA from horizontal cells is required for feedback inhibition of photoreceptors. PMID: 27022629
  7. the deletion of VIAAT in GlyT2-Cre expressing neurons also strongly affects GABAergic transmission PMID: 25027639
  8. This study showed that Conditional Knock-Out of Vesicular GABA Transporter Gene from Starburst Amacrine Cells Reveals the Contributions of Multiple Synaptic Mechanisms Underlying Direction Selectivity in the Retina. PMID: 26400950
  9. Results indicate the significant contribution of not only GAD65, GAD67 and VGAT-mediated GABAergic but also glycinergic transmissions to both palate and abdominal wall formations PMID: 25545713
  10. This study demonstrated that Genetic absence of the vesicular inhibitory amino acid transporter differentially regulates respiratory and locomotor motor neuron development. PMID: 24276495
  11. results provide genetic, behavioral, and electrophysiological evidence that VGAT-mediated inhibitory drive alters very specific forms of sensory processing: those related to pain processing PMID: 22275517
  12. Results suggest that the vesicular GABA transporter VGAT is fundamental for the GABA- and/or glycine-mediated transmission that supports embryonic development. PMID: 21190592
  13. Cleft palate is caused by CNS dysfunction in Gad1 and Viaat knockout mice PMID: 20333300
  14. Viaat is expressed in horizontal cells of the outer retina not only at terminals invaginating into cone pedicles but also at terminals contacting rod spherules. PMID: 12115694
  15. determined the genomic structure of the mVGAT gene and analyzed the promoter activity of the 5'-flanking sequence PMID: 12573541
  16. functional neuronal VIAAT is present in glucagon-containing secretory granules in pancreatic alpha-cells PMID: 12882924
  17. lack of vgat impairs the function of the respiratory network in mouse fetuses and the impairment progresses with fetal age PMID: 17418495
  18. These findings demonstrate that GABA uptake and release via GAT-1, GAT-3, and VGAT are initially established in the inner retina during the first postnatal week and that these systems then mature in the outer retina during the second postnatal week. PMID: 18975268
  19. The vesicular GABA transporter (VGAT) possesses an uneven number of transmembrane domains, with the N terminus facing the cytoplasm and the C terminus residing in the synaptic vesicle lumen. PMID: 19052203
  20. VGLUT1 is sorted diurnally to the plasma membrane to modulate glutamate transmission during a day/night cycle, whereas VGAT expression is not oscillating but is increased in the presence of a light/dark cycle. PMID: 19103593
