To produce the recombinant Musa acuminata UTP--glucose-1-phosphate uridylyltransferase (UGPA), the gene fragment encoding the full-length Musa acuminata UGPA protein (1-467aa) is cloned into a plasmid vector, creating a vector construct. This construct includes a C-terminal 6xHis-tag for purification purposes. The resulting vector construct is then introduced into yeast cells through transfection. Transfected yeast cells are selected using specific antibiotics, and they are subsequently cultured under appropriate conditions. The expression of the recombinant Musa acuminata UGPA protein is induced within the yeast cells using specific inducers. After successful expression, the yeast cells are harvested and lysed, allowing the release of cellular contents, including the recombinant UGPA protein. Purification of the recombinant UGPA protein is carried out from the cell lysate, achieving a purity level exceeding 90% as confirmed by SDS-PAGE analysis.
