The recombinant Musa acuminata UTP--glucose-1-phosphate uridylyltransferase(UGPA) expression in E.coli requires the insertion of the gene of interest (GOI) into a plasmid vector and the transferral of this vector into E.coli. The gene of interest is formed by fusion the 6xHis-tag gene to the C-termius of the DNA fragment that codes for the full-length Musa acuminata UGPA protein (1-467aa). The transfected E.coli cells are then cultured and induced to express the desired recombinant UGPA protein. The cells are harvested by centrifugation and lysed to release the cellular content, including the recombinant UGPA protein. The recombinant UGPA protein is purified from the cell lysate, with up to 90% purity as measured by SDS-PAGE.
