Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
General amino-acid permease involved in the uptake of all the naturally occurring L-amino-acids, related compounds such as ornithine and citrulline, some D-amino acids, toxic amino acid analogs such as azetidine-2-carboxylate, and the polyamines putrescine and spermidine. Senses its transport substrates to set an appropriate level of transporter activity at the cell surface. Required for FLO11 expression and invasive growth.
Gene References into Functions
  1. The results presented here suggest some possible synergistic effects of phosphorylated Alp1, Gap1 and Can1 amino acid permeases on regulation of amino acid utilization and metabolism in Saccharomyces cerevisiae. PMID: 26865023
  2. results support the concept that different substrates bind to partially overlapping binding sites in the same general substrate-binding pocket of Gap1, triggering divergent conformations, resulting in different conformation-induced downstream processes PMID: 24852066
  3. Although the Bul proteins mediate Gap1 ubiquitylation of two possible lysines, Lys-9 and Lys-16, the Aly proteins promote ubiquitylation of the Lys-16 residue only. This stress-induced pathway of Gap1 down-regulation targets other permeases as well. PMID: 24942738
  4. study show that Gap1 and Can1 permeases undergo ubiquitin-dependent endocytosis in response to their substrates and that this endocytosis is due to transport catalysis rather than intracellular substrate accumulation. PMID: 25266656
  5. a possible mechanism involved in the regulation of Rsp5 activity for Gap1 down-regulation via the phosphorylation of a conserved Thr357 in the Nedd4 family is proposed PMID: 23517290
  6. Internal amino acids promote Gap1 permease ubiquitylation via TORC1/Npr1/14-3-3-dependent control of the Bul arrestin-like adaptors. PMID: 22966204
  7. Transport activity-dependent intracellular sorting of the yeast general amino acid permease PMID: 21471002
  8. identified a self-propagating extrachromosomal circular DNA molecule that results from intrachromosomal recombination between long terminal repeats flanking GAP1 PMID: 20937885
  9. In the present study, we found that Gap1p also catalyzed the uptake of putrescine and spermidine, but not spermine. PMID: 15707981
  10. Mutations in LST4 and LST7 genes, previously identified for their role in Gap1p sorting, similarly block multivesicular endosome to plasma membrane trafficking of Gap1p. PMID: 16641373
  11. Together, these studies provide evidence that the GSE complex has a key role in trafficking Gap1p out of the endosome and may serve as coat proteins in this process. PMID: 16732272
  12. Together, our findings uncover a new mechanism for inhibition of permease activity in response to elevated amino acid levels and provide a physiological explanation for the stringent regulation of Gap1p activity in response to amino acids. PMID: 16885415
  13. Coupled biogenesis of Gap1 and sphingolipid (SLs) would create an SL microenvironment essential to the normal conformation, function, and control of ubiquitination of the permease. PMID: 17553927
  14. Gap1p activity is lowered in cells with an activated RAS2(val19) allele or elevated cAMP levels whereas cells with inactive ras2 allele lose ammonia repression of Gap1p-mediated transport PMID: 17919965
  15. different ubiquitin signals act at the Golgi and plasma membrane to direct GAP1 trafficking PMID: 18434603
  16. The mechanism of action of amino acid transport by Gap1, via a ligand-induced specific conformational change that may be part of but does not require the complete transport cycle, is reported. PMID: 19060912
