Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Protein kinase which is essential for the initiation of meiosis and sporulation.
Gene References into Functions
  1. identify the protein kinase Ime2 to bring about Rim4 clearance via phosphorylation of Rim4's intrinsically disordered region PMID: 29738715
  2. Here we show that Ime2 activity is required to induce the translation of SSP2 mRNA at anaphase II PMID: 26246597
  3. Positive auto-regulation of Ime2 produces a bistable system with a normal meiotic state and a more efficient meiotic state. PMID: 23631506
  4. The ectope expression of Ime2 has deterious on cell cycle regulation, toxocity and the response of the DNA damage checkpoint. PMID: 21099355
  5. Epistasis analysis suggests that Ime1 and Ime2 act in the same order in pseudohyphal growth as in meiosis. PMID: 20876298
  6. SCFGrr1p-dependent destruction of Ime2p kinase has a role in glucose inhibition of meiotic DNA replication PMID: 15601864
  7. Rfa2 serine 27 (S27) is required for Ime2-dependent Rfa2 phosphorylation in vivo. PMID: 16294044
  8. plays a role in controlling the exit from meiosis I and a phospho-modification pathway regulates Ime2p during the different phases of meiotic development PMID: 16684773
  9. Histone deacetylase plays a role in regulating the timing of IME2 expression. PMID: 17158929
  10. Ime2 phosphorylates Sum1, a repressor of meiotic gene transcription, on Thr-306 PMID: 17198398
  11. Ime2 kinase activity mediated by the C-terminal domain is required for the efficient production of normal four-spore asci. These data suggest a role for Ime2 in spore number control in S. cerevisiae. PMID: 18339400
  12. The glucose sensors had only a minor role in regulating transcript levels of the two key regulators of meiotic initiation, the Ime1p transcription factor and the Ime2p kinase, but a major role in regulating Ime2p stability. PMID: 18616605
  13. Ime2 is a key activator of Smk1 signaling. PMID: 19087957
  14. Results show that human Cdk2 is a functional homolog for most of Ime2 functions. PMID: 19197163
